摘    要:目的:通过在缺氧条件下体外培养成骨细胞,探讨缺氧对成骨细胞增殖及凋亡的影响。方法:采用酶消化法传代培养乳SD大鼠颅盖骨细胞并建立缺氧模型。用台盼蓝染色法分别计数培养1、3、5、7天的细胞数;用流式细胞仪检测对照组与缺氧1、3、5天的细胞凋亡率。结果:缺氧组的细胞数少于正常氧组;对照组与缺氧1、3、5天的细胞凋亡率逐渐增高。结论:缺氧抑制成骨细胞的增殖,同时促进成骨细胞的凋亡。

Abstract:Objective: To investigate the effects of hypoxia on osteoblasts pr oliferation and apoptosis. Methods: The osteoblasts obtained from the calvaria o f neonatal SD rats were cultured by method of enzyme digestion in vitro. Osteobl asts were counted by trypan blue staining on the 1st, 3rd, 5th, and 7th day afte r culture and the rate of apoptosis was detected by flow cytometer in normal env ironment and on the 1st, 3rd, and 5th day after cultured in hypoxic environment. Results: The cell number of hypoxic group was fewer than that of control group ; the rate of apoptosis increased gradually when the cells were cultured in norm al environment and hypoxic environment for 1?3?5 days. Conclusions: Hypoxia can inhibit the proliferation of osteoblasts and it can also facilitate the apoptosi s of osteoblasts.
