人肝再生增强因子基因转导对大鼠肝硬化的保护作用

摘    要:目的 探讨人肝再生增强因子(augmenter of liverregeneration,ALR)基因转导对肝硬化的保护作用。方法 构建hALR真核表达载体,并以肌肉注射方式转导至硫代乙酰胺诱导的大鼠肝硬化模型中,观察对肝硬化大鼠的代谢支持作用。结果 构建的hALR真核表达载体pchALR肌肉注射后显著降低了血清AST、ALT、ADH水平,延长了生存时间,提高了存活率。结论 人肝再生增强因子基因转导对肝硬化大鼠具有预防和治疗作用,可显著降低血清AST、ALT、ADH水平,延长生存时间,提高存活率。

Abstract:ObjectiveTo investigate the protective effects of human augmenter of liver regeneration (hALR) gene transduction on live cirrhosis in rats. MethodsEukaryotic expression plasmid containing hALR cDNA was purified and injected into the femoral muscle of cirrhotic rats induced by intraperitoneal administration of thioacetamide (TAA) for observation of metabolic support of hALR gene transduction. ResultsIntramuscular administration of hALR plasmid significantly lowered the serum levels of AST, ALT and ADH, prolonged the survival period and increased the survival rate. ConclusionshALR gene transduction exerts preventive and therapeutic effects on cirrhotic rats and can markedly lower serum levels of AST, ALT and ADH, prolong survival period and increase survival rate.
