二氧化氯对细菌杀灭作用机理的实验观察

摘    要:采用悬液定量杀菌试验,紫外分光光度法及电镜观察,研究二氧化氯对大肠杆菌和枯草杆菌黑色变种芽孢的杀灭作用及其作用机理。结果,以含二氧化氯100mg/L溶液对枯草杆菌黑色变种芽孢作用3、5、10、15、30min,杀灭率分别为39.26%、45.20%、88.96%、99.97%、99.99%,DNA漏出率分别为52.73%、56.14%、65.13%、66.79%、69.91%;以含二氧化氯10mg/L溶液对大肠杆菌作用3、5、10、15、30min,杀灭率分别为99.85%、99.92%;、100%、100%、100%,DNA漏出率分别为31.79%、31.87%、32.32%、33.21%、33.57%。在透射电子显微镜下,经二氧化氯作用的枯草杆菌黑色变种芽孢保护性屏障和大肠杆菌细胞壁受抑,通透性发生变化,核心物质漏出。杀灭率和DNA漏出率不成比例关系,说明二氧化氯杀菌还有其他机理。

Abstract:Quantitative suspension germicidal test, ultraviolet spectrophotometry and electronmicroscopy were used to study the efficacy of chlorine dioxide in killing Escherichiu coli and Bacillus subtilis var. niger spores and mechanism of its action. The results showed that the killing rates of Bacillus suhtilis var. niger spores exposed to the solution containing chlo-rine dioxide 100 mg/ L for 3, 5, 10, 15, 30 min were 39.26%, 45.20%, 88.96%, 99.97% and 99.99% respectively and the rates of DNA leakage were 52.73%, 56.14%, 65.13%, 66.79% and 69.91% respectively. The killing rales of Escherichia coli exposed to the solution containing chlorine dioxide 10 mg/L for 3, 5, 10, 15 and 30 min were 99.85%, 99.92%, 100%, 100% and 100% respectively and the rales of DNA leakage were 31.79%, 31.87%, 32.32%, 33.21% and 33.57% respectively. Eleclronmicroscopic examination found that the proteclive barrier of Bacillus subtilis var. niger spores and the cell wall of Escherichia coli were damaged, the permeability changed and the core substance leaked out after exposure to chlorine dioxide. There was no proportional relalion between the killing rate and the rate of DNA leakage, indicaling that there is other mechamism of germicidal aclion of chlorine dioxide.
