盐酸小檗碱对DSS诱导的结肠炎小鼠血小板活化功能的影响

摘    要:目的 :评价盐酸小檗碱对右旋葡聚糖硫酸钠 (DSS)诱导的结肠炎小鼠血小板活化功能的影响。方法 :48只BALB/C雄性小鼠自由饮用8 %DSS的水溶液诱导结肠炎模型后 ,随机分成3组 ,分别接受小檗碱灌胃治疗 (T40组 :40mg/kg/d ,n=16 ;T80 组 :80mg/kg/d ,n=16)或等量蒸馏水灌胃 (MC组 :n=16) ,另外16只未造模雄性小鼠 (NC组 )治疗期间给予等量蒸馏水灌胃 ,每日一次。于治疗3天和7天后各组随机抽取8只处死 ,分别采用放射免疫分析 (RIA)和双抗体夹心免疫放射分析 (IRA)测定血浆TXB2 和P -选择素水平。结果 :小檗碱治疗3天后各组之间血浆TXB2 和P -选择素水平均有显著性差异 (P<0.01) ,治疗7天后各组之间血浆TXB2 和P -选择素水平也均有显著性差异 (P<0.01) ;同一组别治疗3天与7天后相比其水平有显著性差异 (P<0.01)。结论 :小檗碱呈时间、剂量依赖性降低血浆TXB2 和P -选择素水平 ,改善DSS诱导结肠炎小鼠血液高凝状态 ,提示小檗碱可用于UC的治疗。

Abstract:Objective: To evaluate the effects of berberine chloride on platelet activation of mice with dextran sulphate sodium-induced colitis.Methods: After drinking 8% DSS freely for 8 days, colitis was induced in 48 male BALB/C mice, which were then randomized into three groups to receive a daily oral administration of berberine chloride (T40 group:40 mg/kg/day,n=16; T80 group:80 mg/kg/day,n=16) or an equal volume of distilled water (MC group,n=16) for 3 days or 1 week. An additional 16 male mice, which were used as normal control(NC group), drunk water freely instead of DSS, received a daily oral administration of distilled water for 3 days or1 week. Eight mice from every group were killed 3 days and 1 week after the administration of berberine chloride. The levels of plasma TXB2 and P-selectin were measured by RIA and IRA.Results:There are significant differences between the four groups after 3 days or 7 days of the plasma TXB2 and P-selectin levels (P<0.01); The levels of plasma TXB2 and P-selectin in the same group are also different significantly between 3 days and 7 days (P<0.01). Conclusions: Berberine chloride decreases the levels of plasma TXB2 and P-selectin in a time- and dose- dependent manner and improves the procoagulant state of DSS-induced colitis in mice. It suggests that berberine chloride might be used in the therapy of human ulcerative colitis.
