摘    要:目的 研究法尼酯X受体(farnesoid X receptor,FXR)对B类清道夫受体Ⅰ(scavenger receptor class B typeI,SR-BI)表达的调控.方法 FXR配体androsterone不同浓度处理Eahy926细胞,RT-PCR检测FXR特异靶基因小异源二聚体伴侣受体(small heterodimer partner,SHP)表达量的变化.RT-PCR和Real-time PCR分析经androsterone处理后Eahy926细胞中SR-BI mRNA表达的变化.用Western blot检测SR-BI蛋白表达的变化.结果 经androsterone刺激,Eahy926细胞中SHP的表达明显升高,表明FXR被活化.在经androsterone刺激的血管内皮细胞中,SR-BI在转录水平和翻译水平,表达均显著上升.结论 FXR可在血管内皮细胞系Eahy926中上调SR-BI的表达水平.
Abstract:
Objective To investigate the effect of FXR on scavenger receptor class B type Ⅰ(SR-BI) expression.Methods Human vascular endothelium Eahy926 cells were treated with FXR agonist androsterone,and the specific target gene of FXR SHP mRNA was detected by RT-PCR. SR-BI mRNA and protein were determined using RT-PCR, real-time PCR and Westem blotting. Results The level Of SHP mRNA in Eahy926 cells increased after androsterone treatment at different concentrations for 24 h,demonstrating FXR activation in the cells.RT-PCR,real-time PCR and Western blotting detected increased SR-BI expression at both mRNA and protein levels after FXR activation.Conlusion FXR increases the expression of SR-BI in human vascular endothelium cells.
