烧伤皮肤提取物对培养人肺微血管内皮细胞损伤的观察

摘    要:作者采用人肺微血管内皮细胞(EC)培养模型,动态观察了人新鲜皮肤提取物和烧伤皮肤提取物对内皮细胞功能与形态结构的影响。通过对人烧伤皮肤提取物四种不同浓度作用于肺内皮细胞后培养液中,以及不同时相点内皮素-1、一氧化氮、肿瘤坏死因子-α体液因子的检测和镜下细胞形态学的观察发现:(1)用两种烧伤皮肤提取物滴定培养的EC后,其三种因子均异常升高,与对照组及正常皮肤提取物组有显著的差异(P<0.05,或0.01)。(2)各培养组的各时相点组间比较均无显著差异(P>0.05)。(3)形态学观察,以1000μg及2500μg组其细胞形态及结构出现异常变化明显。作者认为,人烧伤皮肤提取物会导致血管内皮细胞培养液中上述三种因子的异常变化。

Abstract:Human lung cultured vascular endothelial cells model was established in vitro. Two kinds of skin extracts (human burned skin extracts, HBSE, and human natural skin extracts, HNSE) were added to cultured endothelial cells. The results showed that HBSE exerted toxic effect on endothelial cells in vitro. There were significantly higher contents of ET-1, NO, TNF-alpha, and abnormal structural changes in EC in the HBSE group.
