苦参碱对大鼠慢性环孢素肾毒性模型TGF-β1和BMP-7表达的影响

摘    要:目的:探讨苦参碱对大鼠慢性CsA肾毒性是否具有保护作用及其可能机制.方法:SD大鼠56只,随机分为对照组(橄榄油0.2 ml/d),CsA模型组(CsA 20 mg·kg^-1·d^-1),安博维组(CsA 20 mg·kg^-1·d^-1+安博维10 mg·kg^-1·d^-1)和苦参碱组(CsA 20 mg·kg^-1·d^-1+苦参碱10 mg·kg^-1·d^-1),实验共4周,于第2周和第4周末2次采集肾脏标本,行常规病理检查,免疫组化检测TGF-β1和BMP-7表达.结果:与对照组比较,CsA组小管间质损伤评分,TGF-β1表达明显增加,而BMP-7明显下降,P<0.05.苦参碱能下调TGF-β1表达,促进BMP-7表达,减轻肾脏小管间质损伤,P<0.05.结论:苦参碱对实验性大鼠慢性CsA肾毒性具有一定的保护作用.

Abstract:Objective: To investigate the renal protective effect and possible mechanism of matrine in experimental cyclosporine A - induced chronic nephrotoxicity, Methods: Fifty six male Sprague - Dawley rats were randomly divided into four groups:control group(olive oil 0.2 ml/d), CsA groap(CsA 20 mg·kg^-1·d^-1), Ab group(CsA 20 mg·kg^-1·d^-1 plus Ab 10 mg·kg^-1· d^- 1 ) ,matrine group(CsA 20 mg· kg^- 1·d^- 1 plus matrine 100 mg· kg^- 1 ·d^- 1 ), with fourteen rats in each group. The rats were sacrificed at the end of weeks 2,4. The kidneys were resected and fixed in 10% neutral formaldehyde for HE staining. Transforming growth factor- beta I(TGF- β1), bone morphogenic protein- 7 (BMP- 7) were determined by irnmunohistochemical method. Results:Compared with controt group, the histologic scores of the tubular- interstitial injury and the expression of TGF- β1 of CsA group were significantly increased, BMP- 7 was significantly lower,P〈0.05. Matrine can dowrtregulate TGF-β1 expression but upregulate BMP- 7 expression in kidney compared with CsA group, P〈 0, 05. Conclusion: Matrine has a renoprotective effect on chronic cyclost:ofine nephrotoxicity rat model.
