摘    要:目的:观察葛根异黄酮对衰老模型大鼠学习记忆能力及海马神经元形态的影响。方法:大鼠随机分为对照组,衰老模型组,药物低、中、高剂量组,采用1%D-半乳糖100mg/kg颈背部皮下注射6周造衰老模型。造模后药物低、中、高剂量组每天分别给予40mg/kg、80mg/kg、160mg/kg葛根异黄酮灌胃给药8周。用药后水迷宫法测试大鼠学习记忆能力,后随将大鼠处死取海马组织,制作石蜡切片,苏木素伊红染色,光学显微镜观察神经元形态结构。结果:用药组与模型组比较,中、高剂量组逃避潜伏期明显缩短、所占距离百分比明显增高;光镜下,模型组与对照组相比锥体细胞明显减少,细胞排列稀疏,细胞核体积明显减少,结构不清楚;药物高剂量组与模型组比较结构、形态基本正常,细胞脱失现象较少。结论:葛根异黄酮对衰老模型大鼠学习记忆能力具有改善作用,对海马神经元具有保护作用。

Abstract:Objective:To study the effects of pueraria isoflavone on learning and memory and changing of neurons morphology in Hippocampi of aging rats induced by D-galactose.Methods:Rats were randomly divided into 5 groups:control group,model group,drug(low、middle、high)groups.The model of aging rats was set up by subcutaneous injecting with 1% D-galactose 100mg/kg into nape for 6 weeks.Drug(low、middle、high)groups were then intragastricly administrated with Pueraria isoflavone(40mg/kg,80 mg/kg,120 mg/kg)for 8 weeks.The learning and memory ability of rats were tested by Morris Water Maze.Then the rats were sacrificed after experiment.The hippocampi were obtained to observing the structure through microscope with HE staining.Results:The escaping latency period in model group was significantly longer than that of pueraria isoflavone(middle、high)groups.The percentage of distance in the platform quadrant in model group obviously decreased compared with the Pueraria isoflavone(middle、high)groups;Comparing with Control group the CA1 pyramidal cell layers of model group were arranged sparse,almost disappeared,the cells nucleus volume was significantly diminished;the structute of hippocampi were not clear.Comparing with Model group,the changes of pueraria isoflavone(middle、high)groups weren't obvious.Conclusion:Pueraria isoflavone could promote the learning and memory ability of aging rats,relieve the damage of neuron and protect the cell.
