塞来昔布对嘌呤氨基核苷诱导的足细胞凋亡中COX-2表达水平的影响

摘    要:目的:旨在通过特异性环氧合酶-2(COX-2)抑制剂塞来昔布对嘌呤氨基核苷诱导的足细胞凋亡中足细胞COX-2表达水平变化的影响,初步探讨特异性COX-2抑制剂对足细胞保护作用的机制。方法:以条件永生性小鼠足细胞为研究对象,分为6组:正常对照组(CON)、嘌呤霉素氨基核苷组(PA)、地塞米松组(DEX)、塞来昔布组(CELE)、地塞米松+嘌呤氨基核苷组(DEX+PA)、塞来昔布+嘌呤氨基核苷组(CELE+PA),分别于0h、8h、24h和48h四个时间点用Hoechst 33258染色检测足细胞凋亡率,24h抽提蛋白用于Western blot检测COX-2的蛋白表达。结果:PA组与正常对照组相比细胞凋亡率明显增多(P<0.001),DEX+PA、CELE+PA组与PA组比较细胞凋亡率明显减少(P<0.05);正常对照组表达COX-2,用PA诱导凋亡后,24h时COX-2表达明显增加,用CELE、DEX干预后COX-2表达水平有所减少(P<0.05)。结论:特异性COX-2抑制剂CELE可以减少PA诱导的足细胞的凋亡率;CELE抑制PA诱导的足细胞凋亡的作用机制可能与CELE对足细胞所表达的COX-2水平变化的影响有一定的相关性。

Effect of Celecoxib on COX-2 Expression of Podocyte Apoptosis Induced by Puromycin Amino-nucleoside

Abstract:Objective:To investigate the direct effect of celecoxib,a specific COX-2 inhibitor,on COX-2 expression of podocyte apoptosis induced by puromycin amino-nucleoside(PA) and establish the basis for studying the protective effects of specific COX-2 inhibitors on podocyte.Methods:The conditionally immortalized mouse podocytes were studied and divided into six groups:Control(CON),puromycin aminonucleoside(PA),dexamethasone(DEX),celecoxib(CELE),dexamethasone+puromycin aminonucleoside(DEX+PA) celecoxib + puromycin aminonucleoside(CELE+PA).Apoptosis was measured by staining with Hoechst 33258 at 0h,8h,24h,48h;Western blot analysis was performed to measure the protein levels of COX-2 at 24h.Results:The percentage of apoptotic cells significantly increased in the podocytes that were exposed to PA compared with control group.In contrast,both dexamethasone and celecoxib significantly reduced PA-induced apoptosis(P0.05).COX-2 expression was seen in control group.It was elevated profoundly after 24 hours induced by PA and reduced by use of DEX and CELE(P0.05).Conclusion:Celecoxib prevents podocyte apoptosis induced by PA.The preventing effect of celecoxib on podocyte apoptosis may be potential interactions with COX-2 expression.
