摘    要:目的探讨抗CD49d单克隆抗体穴McAb雪联用重组人粒细胞集落刺激因子穴rhG-CSF雪动员的外周血干细胞对造血重建的影响。方法经放、化疗预处理的BALB/c小鼠,分别接受经rhG-CSF(实验1组)、抗CD49dMcAb(实验2组)、抗CD49dMcAb联合rhG-CSF(实验3组)动员的同系小鼠的外周血干细胞移植,观察受体小鼠的4周存活率及外周血白细胞(WBC)、骨髓单核细胞(BMNC),粒细胞巨噬细胞集落形成单位(CFU-GM)及脾集落形成单位(CFU-S)等指标。结果实验3组小鼠的4周存活率、WBC、BMNC,CFU-GM、CFU-S数均明显高于实验1和2组,具有显著性差异(P<0.05或P<0.01)。结论抗CD49dMcAb联合rhG-CSF应用能协同动员小鼠外周血干细胞,并能成功重建同系小鼠的造血功能。

Experimental study on hematopoietic reconstruction effects of peripheral blood stem cells mobilized by anti-CD49d monoclonal antibody plus rhG-CSF in mice
Abstract:Objective To study the hematopoietic reconstruction effects of peripheral blood stem cell(PBSC)mobilized by anti-CD49d monoclonal antibody(McAb)with recombinant human granulocyte colony-stimulating factor(rhG-CSF)in mice.Methods BALB/c mice pre-conditioned with chemotherapy and total body irradiation were transplanted with PBSCs from rhG-CSF-mobilized mice(experimental group1),with PBSCs from anti-CD49d McAb-mobilized mice(experimental group2),with PBSCs from above mobilized mice(experimental group3).The white blood cells(WBC)count,four-week survival rate,bone marrow nuclear cells(BMNC),granulocyte-macrophage colony-forming units(CFU-GM),colony-forming unit of spleen(CFU-S)were observed.Results Survival rate,and WBC,BMNC,CFU-GM,CFU-S counts were significantly higher in experimental group3than those of the experimental group1,experimental group2(P<0.01or P<0.05).Conclusions Anti-CD49d McAb and rhG-CSF may synergetically mobilize PBSC in mice.PBSCs mobilized by anti-CD49d McAb combined with rhG-CSF could effectively reconstruct hematopoiesis in mice.
