软肝化坚颗粒对肝纤维化模型C57小鼠肝脏MMP-2和TIMP-2表达的影响

摘    要:目的观察软肝化坚颗粒对肝纤维化模型C57小鼠肝组织中基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶抑制剂-2(TIMP-2)表达的影响。方法 40只C57小鼠被随机分为四组,即正常对照组、模型对照组、软肝化坚颗粒低剂量组和高剂量组,每组10只。采用四氯化碳腹腔注射制造小鼠肝纤维化模型,通过免疫组织化学染色方法检测肝组织中MMP-2和TIMP-2的表达,同时进行阳性表达评分及肝组织纤维化评分(S)。结果造模6周结束后各实验组肝组织MMP-2和TIMP-2表达及纤维化评分与正常对照组比较,差异均有统计学意义(P〈0.05),而MMP-2/TIMP-2比值软肝化坚高、低剂量组与正常对照组比较,差异均无统计学意义(P〉0.05);软肝化坚高、低剂量组肝组织MMP-2和TIMP-2表达和纤维化评分明显低于模型对照组,差异有统计学意义(P〈0.05);且MMP-2和TIMP-2比值也较模型对照组趋于平衡,差异均有统计学意义(P〈0.05)。结论软肝化坚颗粒可通过调节C57小鼠肝组织MMP-2和TIMP-2的表达,降低ECM过度沉积,具有抗肝纤维化功效。

Abstract:Objective To observe the impacts of ruangan huajian granules on the hepatic expressions of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase inhibitor-2(TIMP-2)in C57 mice with experimental hepatic fibrosis.Methods 40 C57 mice were randomly divided into four groups,i.e.a normal control group,a model control group,a low-dose ruangan huajian granules group and a high-dose group,10 mice in each group.The intraperitoneal injection of carbon tetrachloride was used to model hepatic fibrosis for the mice.The immune histochemical staining technique was adopted to detect the expressions of MMP-2 and TIMP-2 in hepatic tissue.Meanwhile,the positive expression score and the hepatic tissue fibrosis score were analyzed.Results In the 6 weeks of modeling,the hepatic tissue MMP-2 and TIMP-2 expressions and the fibrosis score in each experimental group were different significantly in statistics as compared with those in the normal control group(P<0.05).The ratio of MMP-2/TIMP-2 in either the high-dose or the low-dose ruangan huajian granules group was not significantly different as compared with that in the normal control group(P>0.05).The expressions of MMP-2 and TIMP-2 and the fibrosis score in the high-dose and low-dose ruangan huajian granules groups were all obviously lower than those in the model control group,presenting the statistical significant difference(P<0.05).The ratio of MMP-2/TIMP-2 tended to be balanced as compared with the normal control group,indicating the statistical significant difference(P<0.05).Conclusion Ruangan huajian granules modulate the expressions of MMP-2 and TIMP-2 in the hepatic tissue of C57 mice and reduce the excessive deposition of ECM.Therefore this drug is effective on anti-hepatic fibrosis.
