基于网络药理学探究三芪口服液治疗慢性肾小球肾炎的效应机制

目的:研究三芪口服液治疗慢性肾小球肾炎(chronic glomerulonephritis,CGN)的网络药理学机制。方法:根据药物的药物动力学(ADME)特性,通过TCMSP平台筛选以三七、黄芪复方研制的三芪口服液的活性成分,通过Pub Chem平台、Swiss Target Prediction平台进行作用靶点的预测;基于Gene Cards数据库、Dis Ge NET数据库、OMIM数据库检索CGN相关靶点,采用Venny分析工具对药物靶点及疾病靶点进行重叠;借助cytoscape软件构建"活性药物-活性成分-靶点基因"网络,采用String数据库构建蛋白质相互作用网络。并将重叠的靶点信息利用DAVID数据库分析生物过程及通路富集。结果:本研究共获得27个药物活性成分,对应211个靶点,与疾病靶点有关的潜在靶点为42个;主要通过调节激素受体结合、蛋白酪氨酸激酶活性、Rap1信号通路、PI3K-Akt信号通路等发挥治疗CGN治疗作用。结论:三芪口服液通过多靶点-多通路治疗CGN,为解释其效应机制提供理论基础及科学依据。

An Exploration of Therapeutic Mechanism of Sanqi Oral Liquid in Chronic Glomerulonephritis Based on Network Pharmacology

Objective:To investigate the network pharmacology mechanism of Sanqi oral liquid treatment in chronic glomerulonephritis.Methods:We selected active ingredients of Sanqi Oral Liquid which was composed of Sanqi and Huangqi,according to drugs ADME parameters and searched the related targets from the platform of Pub Chem and swiss target prediction.Further,we selected chronic glomerulonephritis related genes from Genecards Databases,Dis Ge NET Databases and OMIM Databases.The Venny platform was employed to match the targets of ingredients and the targets for treating CGN,and then"drug-active compounds-target genes"network was constructed by using Cytoscape software.Finally,the protein interaction network of overlap targets was constructed by using String database,and the analysis of overlap targets’enrichment results of GO and pathway was carried out with the aid of DAVID database.Results:The 27 active ingredients,211 corresponding drug-targets,and 42 disease-related targets were screened out.They mainly play therapeutic roles in chronic glomerulonephritis by regulating hormone receptor binding,protein tyrosine kinase activity,Rap1 signaling pathway,PI3 K-Akt signaling pathway and other pathways.Conclusion:Sanqi oral liquid exert therapeutic treatment for chronic glomerulonephritis through multiple targets and pathways.The present work provides theoretical and scientific basis for explaining its potential mechanism.
