| 摘 要： | 目的检测肝细胞癌组织及肝癌细胞株中PAIP1的表达水平,并分析其表达水平与肝细胞癌的临床病理特征参数的关系。方法采用实时荧光定量PCR(RT-PCR)、WB技术检测新鲜手术标本和肝癌细胞株的PAIP1的表达水平,采用免疫组化方法检测肝癌组织芯片PAIP1蛋白表达水平,比较肝癌组织和癌旁组织PAIP1的表达水平差异,在线工具分析肝癌数据库PAIP1的表达水平与病人临床病理参数的关联性。结果与癌旁组织相比,新鲜标本中癌组织PAIP1 mRNA的表达水平明显增高;PAIP1在肝癌细胞株中高表达;组织芯片中,PAIP1在细胞胞浆中表达,肝癌组织中PAIP1表达水平高于癌旁组织,PAIP1表达水平与与病人生存率呈负相关。结论肝细胞癌中PAIP1高表达,可能在肝细胞癌的发生、发展过程中发挥作用。

| Abstract: | Objective To investigate the expression of PAIP1 in hepatocellular carcinoma tissues and hepatocellular carcinoma cell lines and to explore the association between PAIP1 expressions and clinicopathological features of hepatocellular carcinoma.Methods Expressions of PAIP1 mRNA in hepatocellular carcinoma tissues and hepatocellular carcinoma cell lines were determined by real-time PCR and Western-blotting,PAIP1 protein of paraffin-embedded specimens were examined by immunohistochemistry.The relationship between PAIP1 and prognosis of HCC patients was analyzed by Kaplan-Meier Plotter online database.Results PAIP1 mRNA level was higher in hepatocellular carcinoma tissues and cell lines.A significant overexpression of PAIP1 was found in hepatocellular carcinoma lesions compared to the matching adjacent non-tumor tissues.High expression of PAIP1 was significantly correlated with poor survival rates.Conclusions Both PAIP1 mRNA and protein expressions are higher in HCC tissues.PAIP1 may play a role in hepatocarcinogenesis. |
