EGCG对卵巢癌移植瘤生长及对VEGF和PCNA的影响

摘    要:目的探讨表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对卵巢癌移植瘤生长的影响及可能的机制。方法将卵巢癌裸鼠移植瘤分成5组,给予不同浓度的EGCG(10,30,50 mg·kg^-1)干预,阴性对照组(NC)给予生理盐水(0.2 mL/只),阳性对照给予紫杉醇(Paclitaxel)(5 mg·kg^-1)。肝脏组织进行HE染色。实时荧光定量PCR(quantitative real-time PCR,RT-qPCR)和Western blot方法检测裸鼠移植瘤组织中血管生成因子(vascular endothelial growth factor,VEGF)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的mRNA水平和蛋白水平的表达。结果EGCG可以抑制移植瘤的生长以及VEGF和PCNA的表达(P<0.05)。肝脏切片结果显示,紫杉醇组肝细胞核固缩,肝小叶之间出现明显间隙,而EGCG高剂量(50 mg·kg^-1)组无明显变化。结论EGCG抑制卵巢癌SKOV3细胞裸鼠移植瘤的生长,其可能机制与抑制VEGF和PCNA的表达有关。

Abstract:Aim To explore the effects of EGCG on xenografts of ovarian cancer in nude mice and its possible mechanism.Methods Nude mice xenografts of ovarian cancer SKOV3 cells were established and divided into five groups after tumor formation,in which three groups were given EGCG(10,30,50 mg·kg^-1),the NC group was given normal saline(0.2 mL),and the positive control was given paclitaxel(5 mg·kg^-1).EGCG and normal saline were administered once a day and paclitaxel every three days by intraperitoneal injection.Hematoxylin and eosin(HE)was used to stain the liver tissues.Quantitative Real-time PCR(RT-qPCR)and Western blot were applied to detect the expressions of vascular endothelial growth factor(VEGF)and proliferating cell nuclear antigen(PCNA)in tumor tissues.Results EGCG significantly inhibited the growth of ovarian cancer transplanted tumors and the expression of VEGF and PCNA.HE staining showed that the paclitaxel group had nuclear condensation and obvious interlobular space,but there was no significant change with EGCG(50 mg·kg^-1).Conclusions EGCG inhibits tumor growth in ovarian cancer implanted mice,probably associated with the inhibition of the expression of VEGF and PCNA.
