砖茶氟在高海拔大鼠体内24小时代谢实验观察

摘    要:目的观察砖茶氟在高海拔大鼠体内24 h吸收、分布及排泄的动态变化。方法大鼠分两组,用灌胃方法分别给予氟化钠(NaF)和砖茶水,含氟为300 mg/L,染氟后0.25、0.5、1、2、4、8、16、24 h处死动物,取血清、小肠、股骨、肌肉、心、肝、肺、脾、脑及肾,用氟离子选择电极法测定氟水平。结果两组大鼠骨氟最高,24 h 内呈现明显的蓄积作用,至24 h时,NaF组骨氟增加了约77 mg/kg,砖茶组骨氟增加了约94 mg/kg;血中砖茶氟高峰出现较NaF早,随时间延长,血氟水平逐渐降低,至24 h时,血中NaF和砖茶氟水平仍然很高,可见在 24 h内机体不能将摄入的氟全部排出。两组大鼠各器官不同时间氟水平比较可见,砖茶氟和NaF的代谢趋势基本一致,表现为在大多数器官(脾脏除外)中含氟量峰值均出现在灌胃后0.25 h,然后逐渐下降,至24 h,除骨组织外,两组氟在肌肉、心脏、肝脏、肺脏和肾脏等软组织中也有轻度蓄积作用。结论在高海拔大鼠体内,砖茶氟和NaF的代谢趋势基本一致,不同之处在于砖茶氟在骨组织中水平较高。

Abstract:Objective To investigate 24-hour changes of absorption, distribution and excretion of brick-tea fluoride during 24 hours in altitude rats. Methods Rats were divided into two groups and treated by oral intubation with NaF and brick-tea water, respectively. All the animals were sacrificed at 0.25, 0.5, 1, 2, 4, 8, 16 and 24 h after administration. Blood and small intestines, muscles, femurs, hearts, livers, lungs, spleens, brains and kidneys were collected. The fluoride content of all samples was determined by fluoride ion selective electrode. Results Bone fluoride in two groups was the highest. In 24 h, bone fluoride presented obvious accumulation, and it increased about 77 mg/kg in NaF group, about 94 mg/kg in brick-tea group. The climax of brick-tea fluoride content in serum came earlier than that of NaF. Serum fluoride decreased along with time, but still presented at a high level, which demonstrated that fluoride ingested could not been excreted completely in 24 h. In comparison of fluoride level in different organs and time of two groups, the metabolic trends of brick-tea fluoride and NaF were accordant,which was that fluoride peak in most organs (except spleen) occurred at the 0.25th h, and then gradually decreased, and till the 24th h, except bone tissues, fluoride of two groups in muscle, heart, lung, liver and kidney likely had a slight accumulation. Conclusions The metabolic trends of brick-tea fluoride and NaF in high altitude rats are accordant, except that the content of brick-tea fluoride in bone is higher.
