高糖培养ECV304内ROS含量变化及丹瓜方影响的研究

摘    要:目的:探讨高糖培养人脐静脉内皮细胞(ECV304)内ROS含量的变化及丹瓜方的影响。方法以标准的M199培养基(A组,含糖5.55mmol/L)、高糖培养基(B组,含糖16.67mmol/L)、不同浓度的丹瓜方培养基(在B组基础上:C组含1/150丹瓜方,D组含1/300,E组含1/600)、及1/300丹瓜方培养基(F组,在A组含1/300丹瓜方),培养ECV30472小时,以流式细胞仪(flow eytometry,FCM)测定培养细胞内活性氧簇(ROS)含量。结果B组细胞内ROS含量较A组升高(319.87vs41.78)具有显著统计学意义(P〈0.001),D组及E组细胞内ROS含量均较B组降低(197.13or189.70vs319.87),统计学意义也显著(P〈0.001);F组细胞内ROS含量较A组降低(36.26vs41.78),但尚未达到统计学意义。C组细胞内ROS较B升得更高(581.82vs319.87),统计学意义显著(P〈0.001)。结论:血管内皮细胞经高糖培养可显著升高其胞内ROS含量;适当浓度丹瓜方对高糖培养血管内皮细胞内ROS含量具有显著降低作用。

Abstract:Objective:Research on ROS Content Variance of inner-ECV304 in High Glucose Medium and Inflence on the content of ROS by Dan Gua recipe. Methods: To cultivate endothelial cell of vein (ECV 304) in the following mediums respectively: M199(Group A, glu 5.5mmol/l), high glucose M199(Group B, glu 16.67mmol/l), high glucose M199 Dangua recipe in different density(Group C: 1/150, Group D: 1/300, Group E: 1/600. glu 16.67mmol/l), M199 1/300 dangua recipe(Group F, glu 5.55mmol/l). Cultvated ECV 304 for 72 hours. The content of ROS inner-ECV304 mensurated with flow cytometry (FCM). Results: ROS content in group B rose more than that of group A (319.87 vs 41.78, P<0.001). It descended in group D and group E compared to group B (197.13 or 189.70 vs 319.87,both P<0.001). It also descended in group F compared to group A(36.26 vs 41.78),however P>0.05. Unexpectedly, ROS content in group C rose more than that of group B even(581.82 vs 319.87, P<0.001). Conclusions: ROS content of inner-ECV304 rises significantly while cultivated by means of high glucose medium. Dangua recipe in debita spissitudo has a function of descending ROS content inner-ECV 304 cultivated in high glucose medium. It is a significant cause of dangua recipe in preventing and curing diabetic chrinic complications.
