Abstract:Objective In order to figure out the midgut bacterial community diversity of C. quinquefasciatus in dry season in Hainan Province and deeply reveal the relationship between the host and midgut microbes. Methods In this study, the wild mosquitoes, C. quinquefasciatus were collected in the residential area of Xiqing industrial zone in Dacheng Town, Danzhou City, Hainan Province from March to May. Subsequently, the DNA of midgut bacteria was extracted, and the diversity of midgut bacteria was analyzed by 16S rDNA sequencing. Results At phylum level, proteobacteria was universally captured as the dominant phylum, while the bacteria of phyla Firmicutes, Actinobacteria, Bacteroidetes, Spirochaetes and Verrucomicrobia were also been found in this study. At the order level, Enterobacteria had the highest relative abundance, which was 36.29%. Rickettsiaes, Pseudomonadales, Rhizobiales, Bacillales, Micrococcus all exist in the midgut of Culex quinquefasciatus in the dry season, the relative abundances were 13.77%, 8.05%, 7.12%, 6.56%, and 3.42%, respectively. In addition, genara Pantoea and Wolbachia are the dominant microbes in the samples, and the relatively abundances were 18.42% and 12.64%, respectively. The relative abundances of other bacterial species were less than 10%, including genera Serratia, Staphylococcus, Enterobacter, Acinetobacter, Pseudomonas, and others. Conclusions This investigation reveal the diversity of midgut bacteria of C. quinquefasciatus, the dominant bacterial community of C. quinquefasciatus is captured at the phylum, order and genus level. Timely grasp the changes of midgut microbes of C. quinquefasciatus and study the potential relationship between both of them may provide insight into the biological control of mosquitoes in the future.
