摘 要: | 目的:构建硬皮病小鼠动物模型。方法用100μg/0.1ml 博来霉素每天皮下注射6周龄 BALB/ c 雄性小鼠背部皮肤3周,造模组与空白和 PBS 两组为对照组,各组为10只。取注射局部皮肤进行 HE 染色和 Massion 染色,镜下观察和测定各组皮肤厚度、胶原纤维含量,新鲜皮肤组织测定羟脯氨酸和胶原含量。结果3周后造模组较对照组,皮肤增厚变硬,弹性降低,真皮层明显增厚,胶原纤维明显增粗、膨大,毛囊明显减少,炎性细胞浸润,胶原纤维、羟脯氨酸和胶原含量明显升高。结论符合硬皮病皮肤病理改变,构建硬皮病小鼠动物模型成功。

Abstract: | Objective To establish animal models with scleroderma in mice. Methods Bleomycin was subcutaneously injected into the back skin of male BALB/ c mice aged 6 weeks every day for 3 weeks at the dosage of 100μg/ 0. 1ml. Thirty male BALB/ c mice were divided into model-established group and blank control group and PBS control group, each group of ten mice. skin thickness and collagen fiber content of tissue of the injection of local skin were measured by HE staining and Massion staining. Hydroxyproline and collagen content of fresh skin tissue were detected. Results Compared with the control group after 3 weeks, sclerotic skin in mice of model-established group was present thickening and hardening, the elasticity reduced, the dermis obvious thickening, hair follicles decreased significantly, collagen fibers increases obviously and enlargement, inflammatory cells infiltration and content of collagen fiber and hydroxyproline and collagen increased significantly. Conclusion Animal model in mice with scleroderma can be established successfully in BALB/ c mice after 3-week injection of bleomycin. |
