青蒿琥酯对人前列腺癌PC3细胞的增殖抑制作用及机制探讨

目的探讨青蒿琥酯（Art）对人前列腺癌PC3细胞株增殖的抑制作用及机制。方法用不同浓度Art干预PC3细胞。采用MTT法检测用药后细胞增殖水平；用流式细胞术（FCM）检测细胞周期的改变和凋亡率；采用Western blot法检测不同浓度Art作用48h后PC3细胞Livin和Caspase-3蛋白的表达情况。结果与对照组比较，Art可显著抑制PC3细胞增殖（P〈0．01）。PC3细胞主要被阻滞在G0／G1期，PC3细胞凋亡率各实验组均明显高于对照组（P〈0．01），且呈时间剂量依赖性。与对照组相比，各实验组PC3细胞survivin的表达显著下调（P〈0．01），而Caspase-3的表达增加（P〈0．01）。结论Art可抑制PC3细胞增殖，其作用机制可能与调控细胞周期以及下调Livin的表达、上调Caspase-3而诱导PC3细胞凋亡有关。

Inhibition effect and mechanism of artesunate to PC-3 cell proliferation

Objective To investigate the inhibition effect and mechanism of artesunate（Art） to PC3 cell proliferation. Methods PC3 cell was treated with different coneentrafion Art. And then, the proliferation of PC3 were detected by MTT; the cell ratio in the different stages of the cell cycle and apoptosis ratio of PC3 were detected by flow eytometry（FCM）. After treated with different concentration for 48 hours, Caspase-3 and Livin protein were detected by Western blot. Result Compared with the control group, the proliferation of PC3 was significantly inhibited by Art（ P 〈 0. 01 ） ;the cell cycle of PC3 was mainly blocked at G0/G1 stage; compared with the eontrol group, the apoptosis percentage of the PC3 in every experiment groups were showed a significant high（ P 〈0.01 ） , and Art could inhibit the proliferation and induce apoptosis of PC3 cell in a time-dose dependent manner, compared with the eontrol group, every experiment groups were showed livin protein was remarkablely decreased （P 〈 0.01 ）with different concentrations Art, hut Caspase-3 protein was significantly enhaneed （ P 〈 0.01 ）. Conclusion Art can inhibit the proliferation of PC3 cell, and probable mechanism relate with modulating cell cycle distribution, and inducing PC3 apoptosis through downregulate livin protein and upregulate Caspase,3 protein.
