消渴脂平胶囊对糖尿病大鼠脂肪组织MCP-1影响的研究

摘    要:目的 本研究主要观察消渴脂平胶囊对高脂饲料与链脲佐菌素诱导的糖尿病大鼠模型脂肪组织 MCP-1 表达水平的影响.方法 复制糖尿病大鼠模型分为正常组、模型组、消渴脂平组及马来酸罗格列酮组.灌胃8周后采用免疫组化技术,从蛋白水平观察各组脂肪组织中MCP-1 表达的情况.结果 ①消渴脂平胶囊可降低大鼠FPG及CRP水平.②糖尿病大鼠脂肪组织中MCP-1 蛋白表达情况:正常组脂肪组织MCP-1表达强度较弱、范围小;模型组脂肪组织MCP-1表达显著增强;马来酸罗格列酮组脂肪组织MCP-1表达较模型组减弱;消渴脂平胶囊组脂肪组织MCP-1表达较模型组减弱,与马来酸罗格列酮组接近.半定量分析结果显示:正常组局部弱染,模型组弥漫性染色增强,马来酸罗格列酮局部染色趋强,消渴脂平胶囊组局部弱染,显著低于模型组.结论 ①消渴脂平胶囊具有降糖和改善炎症状态的作用.②糖尿病大鼠脂肪组织中MCP-1 蛋白表达水平升高,提示MCP-1可能是促进肥胖T2DM发生发展的重要炎症因素;消渴脂平胶囊可降低糖尿病大鼠脂肪组织中MCP-1蛋白表达.

Abstract:Objective In this study, lipid levels observed Xiaokezhiping Capsule on high - fat diet and streptozotocin - induced diabetic rat model of adipose tissue MCP-1 expression levels. Methods Diabetic rats were divided into nounal group, model group, Xiaokezhiping capsules group,Rosiglitazone maleate group. After 8 weeks, fed by irrcnunohistochemical techniques,adipose tissue was observed in each group MCP-1 protein expression levels from. Results ①Xiaokezhiping Capsules can reduce FPG and CRP levels in rats;②MCP-1 protein expression of adipose tissue in diabetic rat.. MCP-1 expression of normal adipose tissue has weak intensity,small - scale; Significantly enhanced MCP-1 expression in adipose tissue of molel group; Rosiglitazone maleate group MCP-1 expression in adipose tissue compared with the model group decreased;MCP-1 expression in adipose tissue compared with the model group decreased in Rosiglitazone maleate group; MCP-1 of adipose tissue in Xiaokezhiping Capsule group compared with the model group decreased, close to rosiglitazone maleate group. Semi- quantitative analysis showed that:Its local is stained weakly in normal group, Diffuse stained strengly in model group, Its local is stained strengly in Rosiglitazone maleate group, Local weak staining in in Xiaokezhiping Capsule group was significantly lower than the model group. Conclusion ①Xiaokezhiping Capsule take a role of reducing blood sugar and improving inflammation;②It may be important to promote the development of inflammatory factors in obese T2DM,because MCP-1 protein levels of adipose tissue in diabetic rat increased. Xiaokezhiping Capsule can reduce Capsule MCP-1 protein expression in adipose tissue of diabetic rats.
