卵巢癌血清差异表达蛋白分析

摘    要:目的:探讨卵巢癌血清差异表达蛋白对提高卵巢癌的诊断的意义。方法:采用弱阳离子(WCX)磁珠纯化试剂盒和基质辅助激光解吸电离飞行时间质谱(MALDI—TOF—MS)技术,对41例晚期卵巢癌(FIGOⅢ、Ⅵ期)、40例良胜卵巢肿瘤患者及40例健康人群对照血清标本进行检测,筛选出卵巢癌患者与对照组血清中的差异表达质谱峰,并建立诊断模型;再用此模型对8例早期卵巢癌(FIGOI、Ⅱ期)进行测试,检测其用于早期卵巢癌诊断的可行性。结果:在Mr1000—12000区段,发现卵巢癌与对照组间差异表达蛋白峰8个(P〈0.01或P〈0.05),其中低表达的4个,Mr分别为1457,1857,2202,7761,高表达的4个,Mr分别为:2946,5333,5859,5901。用它们建立成卵巢癌的诊断模型,对8例早期卵巢癌患者进行预测,7例预测准确,准确率为87.5%。结论:MALDI—TOF—MS结合磁珠技术能直接检测出晚期卵巢癌患者血清差异表达蛋白,并且适用于早期卵巢癌患者的诊断,对提高卵巢癌诊断的敏感性和特异性具有一定的临床意义。

Analysis of differential expressed protein from ovarian cancer serum

Abstract:Objective:To study the valae of the differential expressed proteins from ovarian cancer serum for early diagnosis of ovarian cancer. Methods : WCX kit ( Bruker Daltonics GmBH) and matrix - assisted laser desorption/i- onization time - of - fligh - t mass spectrometry technology were used to detect serum samples from 41 patients with advanced ovarian cancer,40 patients with benign ovarian tumor and 40 healthy women for screening differential ex- pressed protein peaks. A diagnostic model was established with these protein peaks. And the nthe diagnostic pattern was applied to 8 patients with early - stage ovarian cancer to investigate the feasibility of diagnosis. Results: In the mass range from 1 000 to 12 000 Da,eight differential expressed protein peaks were screened. Among them,four pro- tein peaks with Mr 1457,1857,2202,7761 were down -regulated and the others with Mr 2946,5333,5859,5901 were up - regulated. A diagnostic pattern consisted of these eight protein peaks was established. The pattern was applied to 8 early - stage ovarian cancer serum samples, and 7 serum samples were successfully predicted with the accuracy of 87.5%. Conclusion : Combination of MALDI - TOF - MS and WCX kit technology can directly screen the differren- tial expressed protein from ovarian cancer serum, and these protein were suitable to diagnosis of early - stage ovarian cancer. They have clinical significance for enhancement of sensitivity and specificity of ovarian cancer diagnosis.
