噻唑烷二酮类对糖尿病大鼠血清P物质、氧化应激和皮肤改变的影响

摘    要:目的观察噻唑烷二酮类对糖尿病大鼠血清P物质、氧化应激炎症因子和皮肤改变的影响。方法用链脲佐菌素(STZ)将10周龄的雄性Wistar大鼠诱导成糖尿病大鼠模型,分为糖尿病胰岛素治疗组(DI组,n=13)、糖尿病罗格列酮治疗组(DR组,n=13)、糖尿病对照组(DC组,n=13),并以正常大鼠作对照(C组,n=13)。分别于第16周抽取各组大鼠心脏血,测定TNF-α、IL-6、C-RP、SOD及P物质的水平,并作皮肤病理观察。结果糖尿病模型各组IL-6、TNF-α和C-RP水平均明显高于正常对照组(P0.05),DI组和DR组与正常对照组比较则无差异;糖尿病组的SOD和P物质水平与正常组相比明显降低(P0.01),胰岛素组和罗格列酮组的SOD和P物质二者与糖尿病对照组比较有显著增高(P0.01,P0.05)。组织学观察结果显示:16周,糖尿病大鼠表皮明显变薄,角质形成细胞层次明显减少,局部为单层排列,真皮内胶原减少,脂肪消失;DI组和DR组大鼠皮肤未出现上述明显的组织学改变。结论噻唑烷二酮类能降低大鼠血清炎性介质、升高SOD和P物质水平,对糖尿病大鼠皮肤早期病变具有防护作用。

Abstract:Objective To study the effects of Thiazolidinediones on serum substance P,oxidative stress inflammatory factors and skin changes in diabetic rats.Methods A diabetic rat model was induced by the streptozotocin(STZ)to 10 week-old male Wistar rats.The rats were divided into insulin-treated diabetic group(DI group,n=13),rosiglitazone treatment diabetic group(DR group,n=13),diabetic control group(DC group,n=13),and normal control(C group)(n=13).Heart blood of rats in each group at the end of 16 weeks were collected to measure level of TNF-α,IL-6,C-RP,SOD,and P substance The skin biopsy were performed for pathological examination.Results The levels of IL-6,TNF-α and C-RP of all the diabetic model groups were significantly higher than those of the normal group(P<0.05),No difference was observed in DI and DR groups compared with the normal control group;the SOD and SP level in diabetic group was significantly lower than that in the normal group(P<0.01),the SOD and SP levels were significantly increased in insulin and rosiglitazone groups compared with Diabetes Group(P<0.01,P<0.05).The skin of pathogenic diabetic rats became obviously thinner with keratinocytes significantly reduced,even single-layer arrangement,reduction of dermal collagen and fat tissue loss in DC group at 16th week.No such histological changes were observed in the skin of diabetic rats from DI and DR groups.Conclusion Thiazolidinediones can reduce serum inflammatory mediators,increased SOD,substance P levels and help prevent change of skin lesions in diabetic rats.
