电针对膝骨关节炎MIA模型大鼠滑膜组织中MMP-3/TIMP-1表达的影响

摘    要:目的:观察电针与塞来昔布治疗大鼠膝骨关节炎后,大鼠滑膜组织中基质金属蛋白酶-3( MMP-3)和基质金属蛋白酶抑制剂-1(TIMP-1)含量的变化。方法:选用SD大鼠40只,采用随机数字表随机分成正常组、模型组、药物组和电针组,每组各10只。通过向SD大鼠左侧关节腔内注射单体碘乙酸钠( MIA)并驱赶大鼠运动来建立膝骨关节炎MIA模型。造模两周后,对电针组取“后三里”穴、“阳陵泉”穴进行电针治疗;对药物组给予塞来昔布灌胃治疗;正常组跟造模组不做任何干预。结果:模型组滑膜组织中MMP-3和TIMP-1的含量较正常组显著提高( P<0.05),但MMP-3和TIMP-1的变化不同步;经过两周的治疗,电针组跟药物组滑膜组织中的MMP-3和TIMP-1含量较造模组显著降低(P<0.05),但仍然略高于正常组。电针治疗组和药物治疗组中两者含量相近(P>0.05),无统计学意义。结论:电针能有效平衡MIA膝骨关节炎模型大鼠中MMP-3和TIMP-1的异常表达,这可能是电针治疗膝骨关节炎有效的机制之一,为针灸治疗膝骨关节炎提供了有力的证据。

Abstract:Objective:To observe the changes of MMP -3 and TIMP-1 expression level in synovium of knee osteoarthritis rats after they were treated with electroacupuncture and oral celecoxib .Methods:Forty cases of male SD rats were randomly assigned into four groups ,and these were normal group , model group , electroacu-puncture group and celecoxib group .KOA model was constructed by injecting mono -iodoacetate ( MIA) to left articular cavity .Electroacupuncture group was treated with electroacupuncture at “Yang-Lingquan”( GB34 ) and “Hou-sanli”( ST36 ) with a needle afflicted to an electroacupuncture apparatus .Celecoxib group was ga-vaged with a dose of 3mg/(kg -1·2ml -1).Model group received no treatment.And normal group was estab-lished as blank .Results:MMP-3 and TIMP-1 in synovium of model group were significantly increased ( P〈0 .05 ) compared with that in the normal group in the fourth week after treatment ,and the balance between them was significantly broken;the contents of MMP -3 and TIMP -1 in electroacupuncture group and celecoxib group were reduced compared with that in the model group ( P〈0.05) and they were back to be balance again after treatment.In addition,there was no great difference between two groups (P 〉0.05).Conclusion:This sduty demonstrates that electroacupuncture can effectively correct the imbalance between MMP -3 and TIMP-1 in synovium of knee osteoarthritis model rats .It may be one of the mechanisms in the treatment of knee osteo-arthritis by electroacupuncture ,which offers powerful support for acupuncture treatment of KOA .
