纳米银温敏型泡沫气雾剂的抗菌活性研究

摘    要:目的考察用于妇女阴道炎的纳米银温敏型泡沫气雾剂的体外抗菌效果。方法大肠埃希菌、铜绿假单胞菌、金黄色葡萄球菌、白色念珠菌作为模型菌,采用微量肉汤稀释法测定制剂的最低抑菌浓度、《中国药典》2010年版抑菌效力试验指导原则测定制剂的抑菌效果,酶标仪测定细菌生长曲线,并评价细菌敏感度。结果纳米银胶体溶液与纳米银温敏型泡沫气雾剂对4种模型菌MIC值相同,说明该制剂不影响纳米银本身的抗菌作用,铜绿假单胞菌MIC值最低,白色念珠菌的抑菌浓度最高,可达到50μg/ml。细菌生长曲线结果也证明上述试验结果,随制剂浓度的升高,抗菌效果增强。制剂浓度低至50μg/ml,28 d后的抑菌作用仍符合药典规定。结论纳米银温敏型泡沫气雾剂对多种菌具有明显抑制效果,有利于进一步的开发研究。

Abstract:Objective To investigate in vitro the anti-bacterial properties of a novel formulation,temperature-sensitive foam aerosol containing silver nanoparticle,which can be used for the treatment of women bacterial vaginosis.Methods The minimal inhibition concentration(MIC) of the aerosol formulation against different strains,E.coli,S.aureus,C.albicans,and P.Aeruginasa,was determined by microplate proliferation test.A microplate reader was used in kinetic mode to detect the growth of bacteria in LB(Luria-Bertani culture medium) media,and the antimicrobial effect of the formulation was determined according to the guiding principles of ChP 2010.Results The MIC values of temperature-sensitive foam aerosol containing silver nanoparticles against four strains of bacteria were similar to those of colloid solution-silver nanoparticles,which demonstrated that the aerosol formulation did not affect the antibacterial activity of silver nanoparticles.The aerosol formulation showed the best inhibition to P.Aeruginosa,and the worst inhibition to C.albicans with the MIC about 50 μg/ml.The antibacterial effect increased with the increase of formulation concentration,and with the concentration as low as 50 μg/ml,the formulation still showed antibacterial effect after 28 d.Conclusion Temperature-sensitive foam aerosol containing silver nanoparticles showed obvious inhibition effects to several strains of bacteria,which effective antibacterial activity is deserved for further development.
