摘    要:脲支原体的定量计数,因其个体微小,一般需要在光学显微镜下计数平板上生长的菌落,得出菌落形成单位(CFU/ml)和在试管中观察液体培养基pH改变,依颜色变化程度确定颜色变化单位(CCU/ml)。本试验用40孔细胞培养板对解脲支原体进行微量CFU和CCU测定,一块细胞培养板相当于40支小试管和5~6块琼脂平板,但只需要1/150~1/10的常规培养基,经比较微量法与常规法差别不大。这种微量的方法容易做到定量规范化、大大减少了器材和培养基的消耗、缩短了其它的辅助工作量。微量法操作简便、误差小、重复性好,并能在同一块细胞培养板上对多份本进行培养和比较。

Abstract:Usually, tiny colonies of ureaplasma urealyticum on the agar plate are counted under microscope for obtaining colony formation unit/ml ( CFU/ml ) and the pH change of the media is determined with color change unit/ml ( CCU/ml ). In' our laboratory, micro-CFU and micro-CCU methods were devised by using a 40 wells culture plate. One plate needs only 1/150-1/10 of the media used in routine. One plate is equivalent to 40 tubes and 5-6 agar plate. There is no statistical difference in result between micro-and routine method. However, ths micro method is esay to manipulate, consumes less media, reproduces well and can perform several samples with the same plate.
