桑抹茶对高尿酸血症模型大鼠血尿酸水平及肠道菌群的影响

摘    要:目的 观察桑抹茶对高尿酸血症模型大鼠血清尿酸及肠道菌群的调节作用。方法 采用高嘌呤饲料喂养复制高尿酸血症大鼠模型,同时给予桑抹茶干预,于实验期末,用全自动生化仪检测血清尿酸、肌酐、尿素氮水平,ELISA法检测血清及肝组织黄嘌呤氧化酶活性和细菌内毒素水平;收集粪便,提取肠道菌群DNA,进行16S rRNA生物信息学分析。结果 桑抹茶干预能显著降低模型大鼠血清尿酸、肌酐、尿素氮水平及血清、肝组织黄嘌呤氧化酶活性;增加肠道乳杆菌比例,降低血清内毒素水平。RAD结果显示,肠道菌群分类水平与血尿酸、黄嘌呤氧化酶活性相关。结论 桑抹茶具有降尿酸作用,该作用可能与调节肠道菌群,降低血清内毒素水平,抑制黄嘌呤氧化酶活性有关。

Abstract:OBJECTIVE To observe the effects of Folium mori Macha (FM) on blood uric acid level and gut microbiota regulation. METHODS Hyperuricemia model was established by feeding SD rats with high-purine diet, rats in experiment groups were orally administered FM at the same time. At the end of experiment, serum levels of uric acid (UA), creatinine (Cr), blood urea nitrogen (BUN) were determined by automatic biochemical analyser and xanthine oxidase (XOD) activities in serum and liver tissue and serum lipopolysaccharide (LPS) levels were determined by ELISA according to the specification of the kits; Faeces was collected and DNA of intestinal microbiota was exeracted then gut microbiota composition was evaluated by 16S rDNA sequencing. RESULTS FM could significantly decrease serum UA, Cr, BUN levels and inhibit XOD activities in both serum and liver tissue. It could increase the ratio of Lactobacillus in intestinal floras and decrease serum LPS levels. RAD analysis showed that the operational taxonomic unit (OTU) level of intestinal floras was related to the level of serum UA and the activities of XOD. CONCLUSION FM possess a function on anti hyperuricemia via regulating intestinal floras, decreasing serum LPS level and inhibiting XOD activities.
