白塞病患者外周血单个核细胞分泌白细胞介素-10干扰素-γ水平的研究

摘    要:目的通过检测白塞病(BD)患者外周血单个核细胞(PBMC)分泌白细胞介素-10(IL-10)、γ-干扰素(IFN-γ)水平,探讨T淋巴细胞亚群与BD发病的关系。方法采用酶联免疫吸附试验(ELISA)测定45例BD患者和20名健康志愿者PBMC培养上清IL-10和IFN-γ水平。结果①BD活动组PBMC自分泌IL-10水平明显升高,与非活动组、对照组比较差异有统计学意义(P<0.01);非活动组明显高于对照组(P<0.01)。②PBMC分泌IFN-γ水平3组(BD活动组、非活动组、对照组)比较差异无统计学意义(P>0.05)。BD患者PBMC分泌IL-10水平明显升高,增高程度与病情活动性相关,IFN-γ分泌水平无明显变化。结论PBMC分泌IL-10水平对BD诊断和病情活动性检测均有一定临床意义。

Abstract:Objective To investigate interleukin-10(IL-10,Th2-type cytokine) and interferon-gamma(IFN-γ,Th1-type cytokine) secreting capacity by peripheral blood mononuclear cells(PBMCs) in 45 inpatients with Behcet′s disease(BD)and 10 healthy individuals,and try to understand the role of Th1/Th2 balance in the pathogenesis of BD.Methods Sandwich ELISA was used to measure IL-10 and IFN-γ level in the supernatant of culture from PBMCs.Results The level of IL-10 produced spontaneously by PBMCs in active stage was significantly higher than that in static phase and healthy person(P<0.01).There are significant difference between active and inactive patients(P<0.01).The IFN-γ level was not statistically different between BD patients and controls(P>0.05).Conclusion IL-10 may play a significant role in the pathogenesis and disease activity of BD.
