CK、Tubulin-β及 PCNA与鼻咽癌放疗后复发与转移的相关性研究

摘    要:目的:探析CK、Tubulin-β及PCNA与鼻咽癌放疗后复发与转移的相关性。方法采用裸鼠体内移植瘤法。根据治疗方式的不同将裸鼠分成放射组与未放射组两组,分别用10只裸鼠接种每组S1、F1细胞。分析鼻咽癌细胞F1、S1放疗前、后的细胞形态、体内增殖及转移能力。结果鼻咽癌及放疗后复发组织中CK、Tubulin-β及PCNA水平与临床分期、颈部淋巴结转移、年龄及性别无关。放疗后复发组织中CK与Tubulin-β及PCNA表达则表现为正相关性( P<0.05)。两组均未发生肺转移,比较放疗后两组F1、S1裸鼠移植瘤瘤体质量,放射组均较未放射组低,且有统计学差异(P<0.05)。放疗后PCNA、CK的表达水平,S1裸鼠移植瘤明显比F1高(P<0.05)。结论鼻咽癌放疗后的复发过程中CK、Tubulin-β的水平与细胞增殖密切相关,但不是鼻咽癌放疗后复发的独立预测指标。

Correlation of CK,Tubulin-beta,PCNA and Recurrence and Transfer of Nasopharyngeal Carcinoma Patients after Radiotherapy
Abstract:Objective To study the correlation of CK,Tubulin-beta,proliferating cell nuclear antigen( PCNA) and re-currence and transfer of nasopharyngeal carcinoma patients after radiotherapy.Methods The method of implanted tumor in nude mice was adopted in this experiment,and were divided into radiation group and non-radiation group.10 mice were vaccinate a-gainst F1,S1 cells of each group.The cellular morphology,proliferation in vivo and transfer ability of nasopharyngeal carcinoma cells F1,S1 before and after radiotherapy were analyzed.Results The levels of CK,Tubulin-beta and PCNA in nasopharyngeal carcinoma and the recurrence tissue after radiotherapy had no correlation with clinical stage,age and sex,cervical lymph node me-tastasis.The CK had positive correlation with the expressions of Tubulin-beta and PCNA in the recurrence tissue (P<0.05).The 2 groups had no lung metastasis after radiotherapy,the transplanted tumor of body mass of F1,S1 in nude mice were compared,the mass in the radiation group were all lighter than those of non-irradiated group,and the differences had statistical significance ( P<0.05 standard).The levels of PCNA,CK in S1 xenografts in nude mice were higher than those in F1 transplantation tumor in nude mice after radiotherapy (P<0.05).Conclusion The levels of CK,Tubulin-beta during the recurrence of nasopharyngeal carci-noma after radiotherapy has close correlation with cell proliferation,but they are not the independent predictor of recurrence after radiotherapy of nasopharyngeal carcinoma.
