摘    要:目的 探讨模拟失重对胃粘膜上皮HFE-145细胞增殖和周期的影响.方法 采用回转器模拟失重,用免疫组化法观察HFE-145细胞PCNA表达的变化,用流式细胞仪测定细胞周期.结果 回转器模拟失重后,与正常对照组相比,回转组HFE-145细胞24、36、48及72 h的PCNA抗体阳性细胞比率无明显差异,12 h回转组阳性细胞比率明显减少(P〈0.05).24、36、48、72 h回转组HFE-145细胞的细胞周期分布与对照组无明显差异,12 h回转组G0+G1期细胞比例明显增多(P〈0.05),S+G2+M期细胞之和显著减少(P〈0.05).结论 回转器模拟失重使胃粘膜上皮HFE-145细胞在12 h时出现了细胞周期阻滞和增殖抑制.

Abstract:Objective To investigate the effects of simulated weightlessness on proliferation and cell cycle of human gastric mucosa cell line HFE-145. Method A rotating clinostat was used to simulated weightlessness. The expressions of proliferating cell nuclear antigen(PCNA) were examined by immunohistochemical stain. The changes of cell cycle were examined by a cytometer. Result Compared with control, the expression of PCNA of cell line HFE-145 showed no significant change after 24 h, 36 h, 48 h and 72 h simulated weightlessness, but decreased significantly after 12 h simulated weightlessness (P<0.05).As compared with control, the cell cycles of cell line HFE-145 in 24 h, 36 h, 48 h and 72 h groups showed no significant change, but the cells in G_0 G_1 phase after 12 h simulated weightlessness increased significantly(P<0.05), and the cells in S G_2 M phase decreased significantly(P<0.05). Conclusion The proliferation of HFE-145 cell is inhibited during simulated weightlessness on a clinostat for 12 h. The conversion of cell from G_1 phase to S phase is also blocked by 12 h simulated weightlessness.
