定志小丸对抑郁模型大鼠齿状回神经干细胞和学习记忆能力的影响

摘    要:目的:观察定志小丸对抑郁模型大鼠海马神经干细胞和学习记忆能力的作用.方法:40只雌性大鼠分为正常对照组、抑郁症模型组、盐水组和定志小丸组.利用Morris水迷宫视频跟踪分析系统评价大鼠学习记忆能力,免疫组织化学方法评价神经干细胞增殖.结果:与正常对照组对比,模型组大鼠的学习记忆能力下降,Nestin阳性细胞数量减少.定志小丸组大鼠学习记忆能力没有明显变化,Nestin阳性细胞数量增加.结论:定志小丸能够对抗慢性应激对学习记忆的损害,可能与其促进神经干细胞增殖有关.

Abstract:Objective:To observe the effect on dingzhixiaowan on the neural stem cells and learning and memory in depression model rats.Methods:40 female Wistar rats were divided into the normal control group,depression model group,saline group and dingzhixiaowan group.The ability of learning and memory were estimated with Morris water maze.Proliferation of neural stem cells was observed with immunohistochemical method.Results:Compared with the normal control group,the ability of learning and memory in the depression group and saline group decreased,and the number of Nestin-positive cells decreased; while the ability of learning and memory in dingzhixiaowan group did not change obviously,and the number of Nestin-positive cells increased obviously.Conclusions:dingzhixiaowan can maintain the ability of learning and memory in depression,and the mechanism is relative to its promoting the proliferation of neural stem cell in the hippocampal formation.
