人参皂甙Rb1对单侧输尿管梗阻大鼠肾脏的抗氧化保护作用

摘    要:目的观察人参皂甙Rb1对单侧输尿管梗阻(UUO)大鼠氧化应激和肾间质纤维化的影响。方法将雄性SD大鼠60只随机分为UUO组、Rb1治疗组、假手术(SOR)组,科素亚(ARB)组。每组15只,术后3、7、14d每组随机选择5只大鼠处死。比色法检测肾脏组织匀浆OH·、MDA、SOD,行HE和Masson染色动态观察肾脏病理学改变。结果与SOR组相比,UUO组大鼠术后3、7、14d,OH·、MDA高于SOR组,SOD低于SOR组。Rb1治疗组大鼠OH·、MDA水平均不同程度低于UUO组,SOD水平均不同程度高于UU0组;其疗效与ARB组相似。与UUO组相比,Rb1能显著减少UUO大鼠胶原在肾间质的沉积,改善肾脏病理损害。相关分析显示,OH·、MDA的表达与间质相对面积呈正相关,SOD的表达与间质相对面积呈负相关。结论人参皂甙Rb1能抗肾间质纤维化;其机制与其抗氧化应激有关。

Antioxidative effects of Ginsenoside Rb1 on rat kidney with unilateral ureteral obstruction

Abstract:Objective To investigate the antioxidative effect of ginsenoside Rb1 on kidney of rats with unilateral ureteral obstruction (UUO). Methods 60 SD male Rats were randomly assigned into unilateral ureteral obstruction model group(UUO), ginsenoside Rbl group (Rb1), sham-operated group (SOR), and ARB-treated group (ARB). At 3,7, 14days after UUO, 5 rats selected randomly from each group were killed. Hydroxy radical(OH· ), malondialdehvde (MDA), and superoxide dinmutase (SOD) in the homogenized lysate of obstructive renal cortex were determined with hughes colorimetry. Renal pathology change was observed dynamically with HE and Masson staining. Results Compared with the SOR group at day 3,7,14 day, the levels of OH· and MDA were increased obviously in the UUO group(P〈 0. 05). Inversely the level of SOD was obviously decreased (P〈0. 05). The levels of OH · . and MDA were lowwer in the Rbl treatment group than UUO group(P〈0.05), Inversely the level of SOD was higher than it. The effect was similar in ARB-treated group and Rb1 treatment group. Compared with the UUO group, Rb1 decreased obviously deposition of rat renal collagen with unilateral ureteral obstruction, and improved renal pathology damage(P〈0.05). The expression levels of OH · and MDA were correlated positively with interstitial opposite area, the expression levels of SOD was correlated negatively with it. Conclusion Ginsenoside Rbl can counteract renal intemtitial fibrosis , and the mechanism is concerned with opposing oxidative stress.
