结核分枝杆菌基因分型及其耐药性分析

摘    要:目的探讨四川地区结核患者结核分枝杆菌基因分型及其与耐药性关系。方法采用PCR和琼脂糖凝胶电泳技术,对124株结核分枝杆菌的6个可变重复位点进行检测,并应用Gel-Proanalyzer3.0软件和BioNumerics(Version3.0)软件对检测结果进行基因型分析,同时分析其基因型与耐药之间的关系。结果124株结核分枝杆菌共分为37个不同的VNTR类型,成簇基因型占74.19%(92/124),单一基因型占25.81%(32/124)。耐药菌株在成簇类型和单一类型的分布上存在统计学差异。结论四川地区结核患者的结核分枝杆菌具有明显的基因多态性,成簇类型是主要流行菌株,应加强流行结核菌株的监控。

A Study on Genotyping of Mycobacterium Tuberculosis and Its Drug Resistance

Abstract:Objective To study on genotyping of Mycobacterium tuberculosis(MTB) isolates and to analysis the relation between drug resistance and genotype of Variable number tandem repeat(VNTR)in Sichuan area.Methods Six tandem repeat loci of the 124 isolates of M.tuberculosis in Sichuan area were analyzed by PCR and agarose gel electrophoresis.The results of genotyping were analyzed with Gel-Pro analyzer 3.1 software and BioNumerics 3.0 software,and the relation between genetic type and drug resistance was explored.Results A total of 37 different allele profiles were identified by the VNTR for all the 124 isolates,74.19%(92/124)and 25.81%(32/124)belonged to cluster type and single type respectively.Drug resistant M.tuberculosis was significantly associated with cluster type.Conclusion There is obvious genetic polymorphism in the M.tuberculosis isolates in Sichuan area.And the clustering type was perhaps the main epidemic strains.It is suggested that surveillance of the epidemic genotype to be strengthened.
