骨髓间充质干细胞对大鼠周围神经端侧吻合后神经再生的影响

摘    要:目的周围神经损伤是临床常见的难题,干细胞移植治疗各种疾病已成为一种新的医学发展趋势。本实验的目的是研究大鼠MSCs移植后对大鼠周围神经端侧吻合后神经吻合口处神经轴突再生的影响。方法 50只Wistar大鼠随机分为2组,均在显微镜下制成腓神经与胫神经端侧吻合模型。1实验组:在端侧吻合术后1周尾静脉注射1×107制备好的MSCs;2对照组:不予特殊处理。分别在术后2、3、4、8、12周进行取材,对比镜下吻合口神经再生情况、测定腓神经功能指数(PFI)、测定双侧胫前肌湿重恢复率。结果术后8周及12周时实验组大鼠行走步态较对照组大鼠稳定;术后8周及12周时实验组大鼠的胫前肌湿重恢复率较对照组明显增加;大鼠神经端侧吻合标本镜下观察:实验组端侧吻合口神经芽突生长较对照组明显,神经细胞也较为丰富。结论周围神经损伤在无条件进行端端吻合修复时可采用端侧吻合的方式进行修复,MSCs可有效地促进端侧吻合后的神经再生,促进神经功能的恢复。

Effect of bone marrow mesenchymal stem cells on the nerve regeneration of peripheral nerve end to side anastomosis of Rats

Abstract: Objective ] Peripheral nerve injury i-s a common clinical problem, there is no effective treatment measures. Stem cell transplantation in the treatment of various diseases has now become a new medical trend. The purpose of this experiment is to study the effect of MSCs transplantation in rats nerve axonal regeneration after peripheral nerve end-to- side anastomosis. Methods ] 50 Wistar rats were randomly divided into two groups , were made to peroneal nerve and tibial nerve end-to-side anastomosis model under the microscope. 1. Experimental group: 1 week after surgery intravenous injection 1 x 107 prepared MSCs; 2. Control group:no special treatment. 2,3,4,8,12 weeks after surgery, respectively, were detected drawn to observe the contrast of lower end anastomosis axonal regeneration under microscope, measured PFI, determination of bilateral tibialis anterior muscle wet weight recovery rate and so on. Results ] After 8 weeks and 12 weeks in the experimental group were compared with control rats gait stability ; experimental rats visible tibialis anterior muscle wet weight recovery rate increased significantly compared with the control group ; rat neural side anastomosis specimens were observed: the experimental group side anastomosis nerve bud outgrowth compared control group , the nerve cells are more abundant. Conclusion ] Peripheral nerve injury repair which were unconditional end-to-end anastomosis, end-to-side anastomosis can be used. MSCs transplanted can effectively promote nerve regeneration of end- to-side anastomosis, and promote recovery of neurological function.
