硬皮病小鼠皮损TGF-β_1 I,III型胶原含量及其相关性分析

摘    要:目的探讨转化生长因子(TGFβ1)在硬皮病发病中的作用及其与COLI,COLIII含量的相关性。方法采用免疫组化技术检测7例硬皮病小鼠模型的皮损中TGFβ1蛋白及I,III型胶原蛋白含量,以7例正常小鼠皮肤组织作为对照(不加BLM的PBS液处置);并对I,III型胶原及TGFβ1的含量进行比较和相关分析。结果TGFβ1及I,III型胶原在硬皮病小鼠皮损的含量均高于对照组(P<0.001);相关分析表明I型胶原的含量与TGFβ1的含量呈正相关(P<0.05)。结论在硬皮病小鼠皮损中有较多的TGFβ1,且硬化皮肤中TGFβ1与I型胶原的含量有显著相关性。提示其在小鼠皮肤硬化过程中起一定作用,可能与硬皮病的病理纤维化的形成有关。

Abstract:Objective To investigate the role of transforming growth factor-β_1 (TGF-β_1) in the pathogenesis of the mouse model of sclerotic skin and to correlation analyze the content of collagen type I (COL-I ), III(COL-III) and TGF-β_1.Methods The Content of TGF-β_1 and collagen type I and III in the skin lesions from 7 mouse models of sclerotic skin ,and skin from 7 normal controls were detected by immunohistochemical method.Results A higher content of TGF-β_1 and collagen type I, III in the skin lesions of the mouse model of scleroderma were seen,compared with those in controls (P<0.001) ,and The direct correlation between the content of TGF-β_1 and the content of collagen type I in the the mouse model of sclerotic skin were found(P<0.05).Conclusion TGF-β_1 is produce intensely in the skin lesions of the mouse model of sclerotic skin,There are direct correlation between the content of TGF-β_1 and the content of collagen type I in the skin lesions of the mouse model of scleroderma. which implies that it may be involved in the pathogenesis of the mouse model of sclerotic skin and may be involved in the pathogenesis of scleroderma.
