摘    要:目的 研究野生天麻的遗传多样性,为合理利用野生天麻种质资源、为天麻的进一步系统选育研究提供一定的理论依据.方法 利用AFLP技术,采用8对M+3和P+3引物组合对23份贵州野生天麻品种进行了总基因组DNA水平上的多态性检测,用NTSYSpc-2.10s 软件的UPGMA 方法 对检测结果 进行聚类.结果 共获得552条可统计的条带,其中396条呈多态性,多态性带百分率约72%.UPGMA聚类可以将贵州不同产地的野生天麻很好地区分开来.结论 该实验揭示了贵州野生天麻种质丰富的遗传多样性,聚类分析结果 表明多数来源地相同的天麻种质表现出较为密切的亲缘关系.

Abstract:Objective To study the genetic polymorphism of wild Gastrodia elata Bl. in Guizhou province, and provide reference for the utilizing its germplasm resources and cultivating new varieties. Methods The phylogentic relationships among 23 wild Gastrodia elata Bl. in Guizhou were analyzed using AFLP to study the genetic diversity with eight AFLP primer combinations (M +3 and P + 3 ) . The determined molecular markers were analyzed through UPGMA method of software NTSYSpc - 2.10 s. Results 552 bands were detected, 396 of which were polymorphic, and the high ratio of polymorphic bands (86% ) were observed in this study. The dendrogram from the clustering of UPGMA showed good classification of all population. Conclusion Our results suggest that wild Gastrodia elata Bl. in Guizhou shows abundant genetic diversities. Results of cluster analysis showed that the classification based on AFLP markers of wild Gastrodia elata Bl. in Guizhou is consistent with their origin.
