血管内皮细胞体外三维培养方法的比较

摘    要:目的 比较不同方法进行脐静脉内皮细胞体外三维培养形成三维血管状结构的差异. 方法 采用表面培养法与混合培养法行脐静脉内皮细胞体外三维培养,观察形成的三维管网状结构. 结果 表面培养法:细胞生长快,管状结构形成多,细胞分支连接形成复杂的三维网状结构;混合培养法:细胞生长缓慢,管状结构形成少,出现细胞连接形成的细胞索与三维网状结构数量较少.两种培养方法形成的血管样结构的数量与长度明显不同. 结论 表面培养法体外血管三维培养,具有操作简便、细胞生长快、三维血管结构形成数量多、血管网络结构复杂等特点,适用于血管生成影响因素的实验研究.

Abstract:Objective To study the difference of two methods of three-dimensional endothelial cell culture in vitro. Methods By using two three-dimensional endothelial cell culture systems, the capillary-like tube-networks formed by the endothelial cells was observed in the surface culture system and the mixing culture system. Results The growth of endothelial cells, the formation of tube-networks and the three-dimensional meshwork by the endothelial cells in the surface culture system was significantly higher than that in the mixing culture system. Conclusion The three-dimensional surface cell culture system is a convenient method which has merit in the growth of endothelial cell, and especially suit for the suitable in the effects of the factors on the angiogenesis.
