杜仲叶绿原酸分离纯化工艺比较及其抗氧化性研究

摘    要:目的:优化杜仲叶中绿原酸的分离纯化工艺,并探讨其抗氧化活性。方法:以绿原酸提取率为指标,比较铅沉淀、石灰乳沉淀、溶剂萃取、壳聚糖絮凝除杂对粗提物的分离效率,并考察7种大孔树脂对绿原酸的吸附性能,还研究了杜仲叶绿原酸提取物的抗氧化活性。结果:最佳分离除杂方法为壳聚糖絮凝除杂法,最佳树脂为NKA-9型大孔树脂,上柱前粗提物绿原酸含量为2.54%,经大孔树脂纯化后产品中绿原酸含量达到42.43%,提高了16倍。结论:壳聚糖絮凝除杂法能很好分离除杂,NKA-9型大孔树脂对杜仲叶绿原酸有较好纯化效果。杜仲叶绿原酸提取物抗氧化活性强于绿原酸标准品。

Abstract:Objective: To optimize the separation and purification method of chlorogenic acid from leaves of Eucommia ulmoides Oliv.and study its antioxidant activities.Methods: The extraction effects of chlorogenic acid from leaves of Eucommia ulmoides Oliv.was used as evaluation index.The optimum separation conditions were designed by using lime precipitation,solvent extraction,precipitation of lead acetate and chitosan to separate chlorogenic acid.Seven types of macroporous resin,including NKA-9,NKA-Ⅱ,D101,XA-8,DA-8,HPD100 and HPD826 were compared the purification efficiency.The antioxidant activity of extracts was evaluated.Results: Chitosan and NKA-9 macroporous resin can be used in the separation and purification of crude extracs of chlorogenic acid from leaves of Eucommia ulmoides Oliv.The chlorogenic acid in stock solution was 2.54%.The purity of the product after the resin purification was 42.43%,which was for increased 16 times.Conclusion: Chitosan and NKA-9 was suitable to separate and purify the chlorogenic acid.The extract of chlorogenic acid from Eucommia ulmoides leaves has more effective antioxidant activity than chlorogenic acid.
