铁皮石斛原球茎悬浮培养研究

摘    要:目的 建立铁皮石斛原球茎悬浮系 ,作为人工种子繁殖体。方法 对铁皮石斛原球茎悬浮培养条件进行比较选择 ,并测定生长增殖状况。结果 悬浮培养的原球茎分散性好、整齐均一。悬浮培养的最适培养基为 1/ 4 MS+40 mg/ L Vc,p H5 .6 ,摇床转速为 5 0 r/ m in,15 d继代一次 ,黑暗培养。培养基中添加 0 .5 m g/ LABA预培养 30 d,原球茎的质量提高、同步性更好。结论 首次报道了铁皮石斛原球茎悬浮系的建立 ,为铁皮石斛大规模培养开创了新途径。

Abstract:Objective To set up a system for suspension culture of Dendrobium candidum used for the explant of artificial seed.Methods The PLBs were culture in different media,condition and factors. The indication of growth and multiplication of the PLBs were studied,and contrasted to the amount,gross weight and net weight of the PLBs. Results The PLBs of supension cultures were finely divided ,well-distrbuted and relatively regular.The best suspension culture medium of PLBs is 1/4MS 40 mg/L Vc , pH5.6, the table shift 50r/min, the medium is changed erery 15 days and cultured in dark.The quality and sunchronization of the PLBs cultured in the medium which were added with 0.5mg/L ABA for 30 days will be improved. Conclusion The suspension culture lines of PLBs of Dendrobium candidum were firstly reported,which opens an alternative road to large-scale culture this herb.
