肿瘤细胞裂解物致敏的树突状细胞对大鼠卵巢上皮癌免疫治疗的体内研究

摘    要:目的:研究树突状细胞(dendritic cells,DC)与肿瘤细胞裂解物致敏的树突状细胞(tumor-associated antigen/dendritic cells,TAA/DC)对荷瘤大鼠的免疫治疗效果。方法:建立大鼠卵巢癌皮下移植瘤模型;于成瘤后2周分别用PBS、DC或TAA/DC进行免疫治疗,比较治疗效果。结果:TAA/DC抑制肿瘤细胞的增殖与转移,对荷瘤大鼠治疗后8周,局部肿瘤无明显增长,P=0.355,无转移灶出现;TAA/DC可明显改善大鼠免疫状况,对荷瘤大鼠治疗后3周,使已经下降的CD8^+ T细胞百分比又明显上升,P=0.000;TAA/DC对荷瘤大鼠单次免疫治疗的效果具有时限性,在对荷瘤大鼠治疗后8周,CD8^+ T细胞百分比再次出现明显下降,P=0.000。结论:TAA/DC可改善荷瘤大鼠的细胞免疫状态,抑制肿瘤细胞的增殖与转移。

Abstract:OBJECTIVE:To study the immunotherapy capacity of DC and TAA/DC to tumor burdened rats in vivo. METHODS: The animal model was established which inoculated with ovarian adenocarcinoma cells. Two-week tumor burdened rats were treated immunologically with PBS, DC and TAA/DC respectively, and the results of the treatment were compared. RESULTS: TAA/DC inhibited the hyperplasia of tumor cell, and made subcutaneous tumor retarded,P=0.355. In TAA/DC treated tumor carring rats, no metastasis was observed. TAA/DC improved the immune condition of the tumor-burdened rats that CD8+ T cells raised remarkably, P=0.000. However,the effect of improved immune condition only sustained a short period, and showed that CD8+T cells reduced before the death,P=0.000. CONCLUSIONS: Specific anti-tumor immunotherapy of TAA/DC could make the immune condition of the tumor carring rat much better. The hyperplasia of tumor cells is inhibited, and the rate of metastasis is reduced.
