摘    要:目的探究miR-125a-5p在非小细胞肺癌(NSCLC)患者血清中的表达情况,分析miR-125a-5p与临床病理特征之间的相关性及其在NSCLC早期诊断中的意义。方法收集2017年7月至2018年3月安徽医科大学附属巢湖医院44例NSCLC患者为试验组和20例体检的健康志愿者为对照组。采用Q-PCR术检测NSCLC患者和健康者外周血清miR-125a-5p的表达量,并评估其临床意义。结果试验组血清miR-125a-5p相对表达量为0. 025(0. 017,0. 051),低于对照组的0. 050(0. 034,0. 117),差异有统计学意义(P <0. 05)。试验组患者miR-125a-5p的表达量与临床TNM分期呈负相关性(r=-0. 688,P <0. 05),而在性别、年龄、吸烟、组织分型间差异无统计学意义,与癌胚抗原(CEA)、细胞角蛋白19片段抗原(CYFRA21-1)水平均无相关性(P> 0. 05)。试验组患者CEA、CYFRA21-1的表达量与临床TNM分期均无相关性(P> 0. 05)。结论 miR-125a-5p在NSCLC患者血清中表达下调,对NSCLC早期诊断优于CEA、CYFRA21-1,可能成为NSCLC早期诊断的重要生物学指标。

Abstract:Objective To investigate the expression of miR-125a-5p in serum of patients with non-small cell lung cancer(NSCLC), and to analyze the correlation between miR-125a-5p and clinicopathological characteristics and its significance in early diagnosisof NSCLC. Methods 44 NSCLC patients in Chaohu Hospital of Anhui Medical University were selected as experimental group and 20 healthy volunteers as control group from July 2017 to March 2018. The expression levels of miR-125a-5p in peripheral blood of the subjects were detected by Q-PCR, and its clinical significance was evaluated. Results The relative expression level of miR-125a-5p in serum of experimental group was 0.025 (0.017,0.051), which was lower than that in control group 0.050(0.034, 0.117), and the difference was statistically significant (P<0.05).The expression of miR-125a-5p in experimental group was negatively correlated with clinical TNM stage (r=-0.688,P<0.05), but not correlated with CEA and cyfra21-1 levels (P>0.05). There was no statistically significant difference in the expression of miR-125a-5p in sex, age, smoking and tissue typing in experimental group. Conclusion MiR-125a-5p is down-regulated in serum of NSCLC patients, which is better than CEA and cyfra21-1 in early diagnosis of NSCLC, and may be an important biological marker for early diagnosis of NSCLC.
