摘    要:目的?建立珍珠母药材HPLC特征图谱,提高其质量控制水平。方法?采用HPLC法测定,Agilent ZORBAX SB-C18色谱柱(4.6?mm×250?mm,5?μm);流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速0.5?mL/min,检测波长254?nm,柱温为30?℃;以苯丙氨酸为参照物,运用国家药典委员会《中药色谱指纹图谱相似度评价系统(2012版)》对测定的10批珍珠母药材数据进行处理分析。结果?建立了以5个特征峰为指标成分的珍珠母药材HPLC特征图谱,以苯丙氨酸色谱峰为参照,其他4个特征峰的相对保留时间分别为0.703±0.006、0.814±0.005、0.881±0.001、1.403±0.008。结论?该方法经过方法学验证,可用于珍珠母药材的质量控制。

Abstract:OBJECTIVE?To establish the specific chromatograms of Margaritifera concha by HPLC, so as to improve its quality control. METHODS?The analysis was performed on an Agilent ZORBAX SB-C18 column (4.6 mm×250 mm,5 μm) with a gradient mobile phase of acetonitrile-0.1% phosphoric acid at a flow rate of 0.5 mL/min. The detection wavelength was set at 254 nm and the column temperature was 30 ℃. Using phenylalanine as the reference solution, 10 batches of samples of Margaritifera concha were analyzed with the developed method,and similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine (2012 Version) was used for the quality assessment. RESULTS?Five common peaks were found through the similarity evaluation system. With phenylalanine as the reference, the relative retention time of the other four peaks were 0.703±0.006, 0.814±0.005, 0.881±0.001, 1.403±0.008, respectively. CONCLUSION?The method has been validated by methodology, and can be used for the quality control of Margaritifera concha.
