丹蛭降糖胶囊对2型糖尿病大鼠瘦素的影响

目的 观察丹蛭降糖胶囊(DJC)对2型糖尿病大鼠血浆瘦素水平的影响.方法 将69只雄性Wistar大鼠随机分为空白组、模型组和4个治疗组(DJC低剂量组、DJC高剂量组、西药组、中西药合用组).空白组喂以普通饲料,模型组与各治疗组喂以高热量饲料,并于4周后腹腔注射小剂量链脲佐菌素制备糖尿病模型.模型制备成功后,空白组和模型组大鼠灌服0.9%氯化钠溶液.DJC低剂量组和高剂量组大鼠分别以540 mg/kg和1 080 mg/kg DJC灌胃.西药组大鼠用马来酸罗格列酮灌胃.中西药合用组大鼠用高剂量DJC联合马来酸罗格列酮灌胃.测定6组大鼠治疗前后的空腹血糖(FBG)和治疗后的血清胰岛素(FINS)、血浆瘦素水平.计算胰岛素敏感指数(ISI).结果 造模后,模型组和各治疗组大鼠FBG水平与空白组比较,差别有统计学意义(P<0.01);治疗后,各治疗组大鼠FBG水平(除外DJC低剂量组)与模型组比较,差别有统计学意义(P<0.05).治疗后,除中西药合用组外,模型组及各治疗组大鼠FINS水平、ISI与空白组比较,差别有统计学意义(P<0.05);除DJC低剂量组外,各治疗组大鼠FINS、ISI与模型组比较,差别有统计学意义(P<0.05).治疗后,除中西药合用组外,其余各治疗组大鼠血浆瘦素水平与空白组大鼠比较,差别有统计学意义(P<0.05);除DJC低剂量组外,其余各治疗组大鼠瘦素水平与模型组比较,差别有统计学意义(P<0.05).结论 DJC能有效改善2型糖尿病大鼠胰岛素抵抗,其机制可能与DJC提高血浆瘦素水平有关.

Effect of Danzhijiangtang Capsules on Leptin in Rats with Type 2 Diabetes Mellitus

Objective To explore the effects of Danzhijiangtang capsules on leptin in rats with type-2 diabetes mellitus(T2DM).Methods Sixty-nine male Wistar rats were randomly divided into blank control group(group A),model control group(group B)and four treatment groups:low-dose Dazhijiangtang group(group C),high-dose group(group D),western medicine group(group E),Dazhijiangtang combined with western medicine group(group F).Group A were fed with ordinary diet,group B and all treatment groups with high-calorie diet.After 4 weeks,T2DM models were prepared by small-dose intraperitoneal injection of streptozotocin.Following preparation,group A and group B were fed with 0 9% sodium chloride solution,groups C and D with Dazhijiangtang capsules(540 mg/kg and 1 080 mg/kg,respectively),group E with rosiglitazone maleate,group F with Dazhijiangtang capsules in combination with rosiglitazone maleate.Pre-and post-treatment fasting blood glucose(FBG)and post-treatment levels of FINS,plasma leptin and insulin sensitivity index(ISI)were determined.Results After preparation,group B and all treatment groups were significantly different from group A in FBG level(P<0.01);after treatment,there was significant difference in FBG level between group B and all treatment groups except group C(P<0.05);group B and all treatment groups except group F differed significantly from group A in FINS and ISI levels(P<0.05);significant difference was noted in FINS and ISI between group B and all treatment groups except group C(P<0.05);groups B,C and D were significantly different from group A in plasma leptin level(P<0.05);significant difference was seen in plasma leptin level between group A and all treatment groups except group F(P<0.05).Conclusion Dazhijiangtang capsules can effectively improve insulin resistance in T2DM rats,the mechanism of which may be related to increase of plasma leptin level.
