膀胱尿路上皮癌中肿瘤转移抑制基因 TIP30/CC3基因的表达及意义

摘    要:目的:探讨膀胱尿路上皮癌肿的肿瘤转移抑制基因 TIP30/ CC3基因的相关表达以及表达的意义。方法收集于2010至2014年存有的完整临床以及病理资料的膀胱尿路上皮癌存档蜡块50例以及癌旁组织50例,通过使用免疫组织化学法对50例膀胱尿路上皮癌存档蜡块以及50例癌旁组织进行相应的检测,分析其中的 TIP30/CC3基因表达水平,同时分析 TIP30/ CC3基因和患者膀胱尿路上皮癌病理特征以及临床特点的相关联系,同时对TIP30/ CC3基因的相关表达进行相应的定量分析,对患者的膀胱尿路上皮癌中以及癌旁组织中的反应阳性颗粒的平均光密度、阳性面积率进行对比。结果 TIP30/ CC3基因在膀胱尿路上皮癌中呈现出一种低表达状态,但在患者的癌旁组织中呈现出高表达状态。癌旁组织中 TIP30/ CC3的平均光密度以及阳性面积率高于膀胱尿路上皮癌,差异有统计学意义( P <0.05);同时 TIP30/ CC3基因和肿瘤的浸润深度以及临床的 UICC 分期之间差异有统计学意义(P <0.05),但与患者的年龄以及性别之间的差异无统计学意义( P >0.05)。结论作为一种抑癌基因,TIP30/ CC3基因在膀胱尿路上皮癌中的低表达情况极有可能促进了膀胱尿路上皮癌的发生以及发展,在临床对膀胱尿路上皮癌患者进行治疗以及预防膀胱尿路上皮癌的过程中需要关注 TIP30/ CC3基因的表达。

Expression and significance of tumor metastasis suppressor gene - TIP30/CC3 gene in bladder urothelium carcinoma
Abstract:Objective To investigate the expression and significance of tumor metastasis suppressor gene - TIP30 /CC3 gene in bladder urothelium carcinoma. Methods Fifty cases of bladder urothelial carcinoma paraffin blocks and 50 cases of carcinoma adjacent tissues paraffin blocks that were stored in Department of Pathology of our hospital from 2010 to 2014 were collected in the study,with complete clinical and pathological data. The expression levels of TIP30 / CC3 gene in these specimens were detected by immunohistochemistry(SP),at the same time,the correlation between the expressions of TIP30 /CC3 gene and pathological characteristics as well as clinical features of bladder urothelial carcinoma was analyzed. Besides the expressions of TIP30 / CC3 gene were quantitatively analyzed,and the average optical density and positive area rate in positive reaction granules were measured and compared between bladder urothelial carcinoma tissues and carcinoma adjacent tissues. Results The low - expression of TIP30 / CC3 gene was observed in bladder urothelial carcinoma,however,high - expression of TIP30 / CC3 gene was observed in carcinoma adjacent tissues. The average optical density and positive area rate in carcinoma adjacent tissues were significantly higher than those in bladder urothelial carcinoma tissues,moreover,there was a significant difference between the expression of TIP30 / CC3 gene and tumor’s infiltration depth as well as clinical UICC stages( P <0. 05). However there was no significant difference between the expression of TIP30 / CC3 gene and patient’s age and sex (P > 0. 05). Conclusion As a tumor suppressor gene,the low - expression of TIP30 / CC3 gene in bladder urothelial carcinoma may promote the pathogenesis and development of bladder urothelial carcinoma,therefore,we should pay much attention to the expression of TIP30 / CC3 gene in treatment and prevention of bladder urothelial carcinoma.
