Abstract:Objective To establish the transplanted tumor producing HCV Cprotein in mice and study the therapeutic effect of DNA vaccine on HCV infection. Methods The recombinant plasmid pcDNA-HCV-C containing HCV C gene was transfected into mouse myeloma Sp2/0 cells with LipofectAMINE. The transfected Sp2/0 cells were cultured in the selective medium containing G418 (400μg/ml) and the Sp2/0-HCV-C cells expressing HCV C antigen were subcutaneously injected into BALB/c mice. Three days later, the inoculated mice were treated with different agents-pcDNA-HCV-C and pcDNA3 and saline as control. Results The tumorigenicity of Sp2/0-HCV-C cells was significantly decreased after treatment with pcDNA-HCV-C as compared with parental tumor cells and mock vector transfected Sp2/0 cells. Moreover, application of the pcDNA-HCV-C plasmid could prolong the survival time of mice. Conclusion DNA vaccination might function as a candidate biotherapy for the treatment of human HCV infectious.
