血脑屏障内皮细胞粘附分子表达与癫痫发病关系

摘    要:目的研究血脑屏障内皮细胞上细胞间粘附分子-1(ICAM-1)表达与癫癎发病的关系.方法用免疫细胞化学法,观察Wistar大鼠皮质和海马区ICAM-1免疫反应阳性表达.结果对照组大鼠皮质、海马区血脑屏障内皮细胞及神经细胞上均有少量ICAM-l免疫反应阳性表达;美解眠致癎组大鼠,上述脑区ICAM-1免疫反应阳性表达显著增强(P<0.01);致癎前给予地塞米松组大鼠,相应脑区ICAM-I免疫反应阳性表达与对照组比较无显著差异(P>0.05).结论表达于血脑屏障内皮细胞及神经细胞上的ICAM-1通过影响血脑屏障通透性及调节脑内免疫反应参与癫癎发病.糖皮质激素治疗癫癎机制之一与其抗粘附效应有关.

Abstract:Aim: To study the expression of intercellular adhesion molecule-1(ICAM-1) on endothelial cells of blood brain barrier (BBB) and pathogenesis of epilepsy. Methods:The expression of ICAM-1 immunoreactivity positive on endothelial cells and neurons in cortex and hippocampus of Wistar rats was observed with immunocytomistry. Results: There was few expression of ICAM-1 immunoreactivity positive on endothelial cells of BBB and neurons in cortex and hippocampus of the control group rats. The expression of ICAM-1 immunoreactivity positive on endothelial cells and neurons in cortex and hippocampus was significantly increased when the rats were induced by bemegride ( P < 0.01). There was no significantly difference in expression of ICAM-1 immunoreactivity positive on endothelial cells and neurons in corresponding areas of rats between the control and administerationdexamethasone before epilepsy group (P > 0.05). Conclusion: The ICAM-1 expressing on endothelial cells and neurons were involved in the pathogenesis of epilepsy by affecting the permeability of BBB and regulating the immune response within brain. One of glucocorticoid mechanisms in treating epilepsy has some relation to its antiadhesion.
