摘    要:目的探讨急性脑梗死患者外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中细胞因子信号转导抑制因子-3(SOCS-3)mRNA和蛋白质表达的变化。方法实时荧光定量PCR法检测60例脑梗死患者和40例健康对照者的PBMC中SOCS-3mRNA表达,Western—blot法检测其蛋白质水平的表达。结果脑梗死患者发病后1~6dPBMC中SOCS-3 mRNA表达明显高于正常对照组,其表达随时间延长呈下降趋势,至第7、14天时与正常对照组无显著差异。其蛋白质的表达在发病后第2-3天升高并达到高峰,之后表达逐渐减少,至第7天仍高于正常水平,第14天降至正常水平。结论急性脑梗死患者PBMC中SOCS-3可能参与了脑梗死后炎症反应的病理生理过程。

Dynamic Changes of Suppressor of Cytokine Signaling-3 (SOCS-3) in Peripheral Blood Mononuclear Cells in Patients with Cerebral Infarcts

Abstract:Objective To detect the expression of suppressor of cytokine signaling-3(SOCS-3) in PBMC from patients suffering from acute phase of cerebral ischaemic stroke. Method The expression of SOCS-3 mRNA in PBMC was determined using real-time PCR from 60 patients sufferring from cerebral infarcts and 40 healthy controls. The protein levels of SOCS-3 were further analysed by Western-blot. Result Compared with the control subjects, the levels of SOCS-3 mRNA from the day-1 to day-6 after the disease onset were significantly increased (P〈0.05). The expression reached a maximum level within 24 hours after the disease onset, and then decreased gradually. From day-7 to day- 14, the expression levels were similar to the control group. The protein levels of SOCS-3 increased rapidly and reached the peak between the day-2 and day-3, and then decreased thereafter. The protein level at day-7 was still higher than the control group (P〈O.05), and the level returned to the baseline level at day-14. Conclusion The expression of SOCS-3 in PBMC was correlated with the pathological development of ischemic inflammation after cerebral infarction.
