三种直接胆红素检测系统的性能评价

摘    要:目的对钒酸盐氧化法、改良J-G法和3,5-二氯苯重氮四氟硼酸盐法(3,5-DCB-4FB)检测直接胆红素(DBil)进行性能评价,探讨3,5-DCB-4FB法是否适用于DBil检测。方法用钒酸氧化法、改良J-G法和3,5-DCB-4FB法分别测定血清DBil,探讨3种方法的精密度、线性、敏感性、抗干扰性和回收率。并分别与Doumas法(手工)进行比较。结果 3种方法平均精密度均在1%~5%之间;平均回收率为103.28%、99.04%、100.69%;线性范围(敏感性)为1.06~335.10μmol/L(1.06)、1.01~310.00μmol/L(1.01)、0.76~310.00μmol/L(0.76)。钒酸盐氧化法抗血红蛋白(Hb)干扰能力优于后2种方法;3种方法与Doumas法进行比较具有良好相关性(r2〉0.975),医学决定水平处的系统误差(SE)分别为-9%、-7%、-1.1%。结论 3,5-DCB-4FB法具有良好的精密度、敏感性、抗干扰能力、线性范围和相关性,与改良J-G法具有很好的一致性。3,5-DCB-4FB法适用于临床常规检测。

Abstract:Objective To evaluate the performance of 3 kinds of direct bilirubin(DBil) detection system vanadic acid oxidation method,improved J-G method and 3,5-dichlorobenzene diazonium tetrafluoroborate(3,5-DCB-4FB) method,and analyze the applicability of 3,5-DCB-4FB in DBil determination.Methods The serum DBil was determined by vanadic acid oxidation method,improved J-G method and 3,5-DCB-4FB method.The precision,linearity,sensitivity,anti-interference ability and recovery were analyzed.The results were compared with those of Doumas method.Results The average precisions were from 1% to 5%.The average recoveries were 103.28%,99.04% and 100.69%.The linear ranges(sensitivities) were 1.06-335.10 μmol/L(1.06),1.01-310.00 μmol/L(1.01) and 0.76-310.00 μmol/L(0.76).The anti-interference ability of hemoglobin(Hb) of vanadic acid oxidation method was better than those of the other methods.3 kinds of detection system compared with the Doumas method had a good correlation(r 2 0.975).The levels of systematic error(SE) were-9%,-7% and-1.1%.Conclusions 3,5-DCB-4FB method has a good precision,sensitivity,anti-interference ability,the linear range and relevance.The 3,5-DCB-4FB method has a good consistency with the improved J-G method.3,5-DCB-4FB method is applicable to routine clinical testing.
