四氧嘧啶建立糖尿病妊娠大鼠模型的研究

摘    要:目的 探讨四氧嘧啶制备糖尿病妊娠大鼠动物模型中不同给药剂量和途径对糖尿病妊娠大鼠动物模型的影响.方法 通过对60例妊娠大鼠尾静脉注射或腹腔注射不同剂量四氧嘧啶,观察成模动物模型率、转阴率和死亡率.结果 除腹腔组(100 mg/kg),其余各组给药前后空腹血糖比较均有显著差异.静脉注射组较腹腔注射组动物模型成模率高、死亡率低;静脉注射组随剂量增加, 转阴率降低,死亡率增加;静脉组2(40mg/kg)的成模率为91.7%,死亡率8.3%,转阴率为9.1%.结论 制备糖尿病妊娠大鼠动物模型四氧嘧啶静脉注射组优于腹腔注射组;40mg/kg静脉注射给药是糖尿病妊娠大鼠动物模型的较好途径.

Abstract:Objective To investigate effects of varied doses and administrations of alloxan on diabetes mellitus models of pregnant mice.Methods 60 pregnant mice were divided into five groups.Three different doses of alloxan were given through caudal vein injection and two different doses of alloxan were given through abdominal cavity injection.The successful modeling rate,urine glucose negative-converting rate,mortality rate were observed for consecutive 18 days and the blood glucose level of pregnant model mice in each group was compared with that of normal pregnant mice.Results The successful modeling rate is higher,and the mortality rate was lower in the caudal vein injection group than those in the abdominal cavity injection group.In the caudal vein injection group,with increase of dose of alloxan,the urine glucose negative-converting rate was decreased and the mortality rate was increased dramatically.Conclusion In establishment of diabetes mellitus in pregnant mice with alloxan,caudal vein injection method is superior to abdominal cavity injection method.40mg/kg of alloxan administered through caudal vein injection for pregnant mice is an more appropriate way.
