川芎嗪对宫内生长受限幼鼠生长发育及脑损伤GAP-43蛋白表达的影响

摘    要:目的:研究钳夹子宫血管致宫内生长受限幼鼠(FGR)脑神经相关生长蛋白-43(GAP-43)的表达及川芎嗪对FGR幼鼠及脑海马GAP-43蛋白表达的影响。方法:通过钳夹子宫血管30 min建立FGR模型,测出生当天及生后7 d假手术组、模型组、川芎嗪低剂量组及川芎嗪高剂量组幼鼠的体重、脑重、肝重,应用免疫组化方法观察川芎嗪对FGR幼鼠脑海马GAP-43蛋白表达的影响。结果:出生当天,模型组幼鼠的体重、脑重、肝重、GAP-43蛋白的表达均明显低于假手术组(P<0.01);川芎嗪低剂量组体重、脑重、肝重、GAP-43蛋白的表达与模型组无显著差别(P>0.05),川芎嗪高剂量组体重、脑重、肝重、GAP-43蛋白的表达明显高于模型组(P<0.01)。生后7 d,4组幼鼠体重无明显差异(P>0.05),但模型组的脑重、肝重及GAP-43蛋白的表达仍显著低于假手术组及川芎嗪高剂量组(P<0.05);同川芎嗪低剂量组无显著差异。结论:足够剂量的川芎嗪对钳夹子宫血管致FGR幼鼠脑组织有明显的保护作用。

Abstract:Objective: To investigate the effect of the tetramethylpyrazine on the expression of GAP-43 protein in the hippocampus of fetal growth restriction(FGR) rats.Methods: Animal models of FGR were established by clamping the uterine vessel of pregnant rats for 30 minutes at gestational 17 d after pre-treatment of tetramethylpyrazine.Body,brain and liver were weighted at birth and on the seventh day after birth.GAP-43 proteins were also measured by immunohistochemistry in the hippocampus.Results: Statistical analysis demonstrated a significant decrease in the weight of fetus,brain,liver and the expression of GAP-43 protein in model group compared with the sham operated group(all P<0.01).No significant difference of the weight of fetus body,brain,liver and the expression of GAP-43 protein was found between the low dose group and the model group(all P>0.05).The expression of GAP-43 protein was increased obviously in high dose group(P<0.01)compared with the model group.The weight of brain and liver of the fetus and the expression of GAP-43 protein in model group were less or lower than in sham operated group and high dose group(all P<0.05) on day 7 after birth.There is no significant difference between model group and low dose group in the above indices(all P>0.05).Conclusion: Higher dose of tetramethylpyrazine has protective effects on brain tissue of FGR rats.
