新型复合纳米银的热凝型丙烯类软衬基托修复材料生物学相容性评价

摘    要:目的:探讨复合纳米银的热凝型丙烯类口腔修复用树脂基托软衬材料的生物安全性,为新型口腔修复材料的开发与临床应用提供安全性参考。方法①复合纳米银的口腔修复软衬材料的制备及使用含10%FBS的正常α-MEM培养液提取材料浸提液。②镜下观察含有浸提液的α-MEM与正常α-MEM培养的牙周膜干细胞的形态及生长状态差异,并比较两组细胞体外的增殖、分化能力。③经皮下注射浸提液及生理盐水后比较两组间有无迟发型超敏反应。④将C57小鼠随机分为浸提液注射组和生理盐水注射组,分别经腹腔内注射纳米银软衬材料浸提液及生理盐水后对小鼠的心、肺、肝、脾、肾取材进行组织学检测。结果①牙周膜干细胞在材料浸提液中状态良好,生长动力学检测(MTT)与对照组比较差异无统计学意义(P〉0.05);两组间成骨、成脂分化能力比较,虽然实验组较对照组呈现略有下降趋势,但最终定量分析发现差异无统计学意义(P〉0.05)。②腹腔内注射实验表明,材料的生物相容性好,无毒性及致敏性。结论复合纳米银的软衬基托修复材料在口腔应用中具有良好的生物学相容性,将有望在临床中获得广泛应用。

Abstract:Objective To investigate the biological safety of nano-silver added to Prosthodontics Underpinning soft lining material,and provide reference for the development and clinical application of new dental prosthetic material to provide security. Methods ①Add nano silver Prosthodontics soft lining material preparation and use of the normalα-MEM culture medium containing 10% FBS extract mate-rial extracts. ②The morphology,growth status,proliferation and differentiation capacity of α-MEM and normal α-MEM cultured periodontal ligament stem cells in vitro were compared. ③The delayed-type hypersensitivity between the two groups after subcutaneous injection of extracts and saline was observed. ④C57 mice were randomly divided into the extraction liquid injection group( n=6 )and saline injection group(n=6),after intraperitoneal injection of nano-silver soft lining materials extracts and saline,the mouse heart,lung,liver,spleen,kidney were taken for histological detection. Results ①In material extracts periodontal ligament stem cells Performance good condition,there was no significant difference compared with the control group in the detection of growth kinetics(MTT)(P〉0. 05). There was no significant difference in the quantitative analysis of estrogenic and adipogenic differentiation ability between the two groups( P〉0. 05 ). ②Intraperitoneal injection experiments showed that the material has good biocompatible,no toxicity and allergenicity. Conclusions Add nano-silver soft liner denture base repair material has good biological compatibility in application,and wide range of applications are expected to be obtained in clinics.
